CD28 ligation induces transplantation tolerance by IFN-gamma-dependent depletion of T cells that recognize alloantigens.

CD28-specific Ab (37.51) has been used in experimental models to mimic the natural ligands and provide costimulatory signals to T cells, thus preventing T cell anergy in vitro (2). Administration of anti-CD28 mAb 37.51 inhibits donor T cell expansion and prevents GVHD in mice (20). To test whether anti-CD28 treatment would prevent GVHD mediated by CD4 and CD8 T cells, a well-established murine GVHD model with C57BL/6 (B6) (H2 b ) donors and B10.BR (H2 k ) recipients was used. Anti-CD28 treatment significantly reduced GVHD lethality compared with control treatment ( P < 0.0001). In allogeneic BALB/c recipients, both WT and CD28-deficient donor (H2 b+ /CD8 + ) cells divided rapidly and expanded extensively, but there was a dramatic difference in the percentage of WT cells with or without anti-CD28 treatment.

Therapeutic effect of anti-OX40L and anti-TNF-alpha MAbs in a murine model of chronic colitis.

Administration of anti-OX40L MAb markedly ameliorated the clinical and histopathological disease in preventive and therapeutic protocols. In vivo treatment with anti-OX40L MAb decreased CD4+ T cell infiltration in the colon and suppressed IFN-γ, IL-2, and TNF-α production by lamina propria CD4+ T cells. The combination with anti-TNF-α MAb further improved the therapeutic effect by abolishing IFN-γ, IL-2, and TNF-α production by lamina propria CD4+ T cells. Histological examination showed prominent epithelial hyperplasia with glandular elongation with a massive infiltration of mononuclear cells in the lamina propria of the colon from the control IgG-treated mice. CD4+ LPL were isolated from the control rat IgG- or anti-OX40L MAb-treated mice at 7 wk after CD4+ CD45RB high T cell transfer and stimulated with anti-CD3 and anti-CD28 MAbs for 48 h.

B7-1 engagement of cytotoxic T lymphocyte antigen 4 inhibits T cell activation in the absence of CD28.

Primed T cells from both types of mice produced cytokines and proliferated in response to stimulator cells lacking B7 expression. However, whereas the response of CD28 1 / 1 T cells was augmented by costimulation with B7-1, the response of the CD28 2 / 2 T cells was strongly inhibited. This inhibition was reversed by monoclonal antibody against B7-1 or CTLA4. To determine whether expression of CTLA4 could indeed be induced in 2C/Rag/CD28 2 / 2 T cells, flow cytometric analysis was performed after restimulation in vitro with P815 cells. As shown in Fig. 2 A, primed CD28 1 / 1 T cells proliferated in response to P815, with a peak response observed on day 2.

The mAb 37.51(anti-mouse CD28 mAb), directed against murine CD28, was purchased from.... Cells (1 x 10^6) were incubated with mAb 37.51 (1 pg/ml) or an irrelevant hamster mAb for 30 min on ice in 100 pl PBS. All cell proliferation tests were performed in 96-well flat-bottom microtiter plates (Nunc, Wiesbaden, FRG), using Click’s/RPMI medium containing 10% FCS and supplements, as described elsewhere. In a first set of experiments, we tested for the expression of CD28 on the Th1 cells used in this study. Next, we tested whether stimulation with anti-CD28 mAb changed the reactivity of Th1 cells blocked by treatment with solid-phase-coupled anti-CD3.

The mAb 37.51, directed against murine CD28, was used to provide costimulatory signals to Th2 cells. CD28 ligation enhanced IL-4 responsiveness in Th2 cells. In vivo administration of anti-CD28 mAb augmented Th2 responses in mouse models. The combination of anti-CD3 and anti-CD28 mAb induced high levels of IL-4 production. This costimulation was essential for the development of Th2 phenotype in vivo.
