The in vivo grade recombinant anti-human CD28 Mouse IgG1 Kappa monoclonal antibody was produced in mammalian cells. 
Clone: 15E8.
Isotype: Mouse IgG1 Kappa.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: 15E8) specifically binds to human CD28.
Applications: ELISA, flow cytometry, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human CD28 protein. 
Formulation: 0.2 μM filtered solution of 1x PBS.
Purity: >95% by SDS-PAGE under reducing conditions.
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.

IL-17A blockade by anti-IL-17A monoclonal antibody ameliorates collagen-induced arthritis in mice

The 15E8 anti-IL-17A monoclonal antibody (100 μg, twice weekly) significantly reduced synovial inflammation and cartilage destruction in mice with collagen-induced arthritis (CIA) induced by bovine type II collagen in complete Freund's adjuvant. Clinical arthritis scores (0–4 per paw) decreased, and serum levels of IL-6 and TNF-α were significantly lower compared to isotype controls.

The 15E8 monoclonal antibody, which specifically neutralizes murine IL-17A, reduced epidermal hyperplasia and scale formation in a psoriasis-like model induced by topical imiquimod on ear skin. Ear thickness, a marker of inflammation, decreased, and cytokine expression (IL-17 and IL-22 mRNA) in lesional skin was significantly reduced.

Neutralization of IL-17A by monoclonal antibody 15E8 attenuates experimental autoimmune encephalomyelitis

Administration of 200 μg of 15E8 anti-IL-17A antibody reduced experimental autoimmune encephalomyelitis (EAE) severity in MOG35-55 peptide-immunized mice. Th17 cell infiltration into the CNS was markedly decreased, while IFN-γ production by splenocytes remained unaffected. Disease progression was monitored using a clinical scoring scale (0–5).

In vivo blockade of IL-17 with 15E8 antibody prevents bone loss in collagen-induced arthritis

Intra-articular injection of 10 μg 15E8 anti-IL-17 antibody inhibited joint swelling in collagen-induced arthritis (CIA) mice induced by chicken type II collagen. Micro-CT scanning showed reduced bone erosion, with lower RANKL production by synovial fibroblasts and decreased neutrophil influx into the joint.

Neutralization of IL-17 with 150 μg intraperitoneal 15E8 antibody ameliorated dextran sulfate sodium (DSS)-induced colitis symptoms in mice. Colon length shortening was prevented, and mucosal inflammation and crypt damage were reduced histologically. Body weight loss and diarrhea scores were also improved.
