MB130: Probe-Based Multiplex One-Step qRT-PCR Kit

The probe-based multiplex one-step qRT-PCR kit is ready-to-use 5x concentrated solution for multiplex, quantitative, reverse transcription-qPCR. Except DNA template and primers, the 5x probe-based multiplex one-step qRT-PCR master mix contains all other components for RT and qPCR, including dNTP/dUTP mix, MgCl2, reaction buffer, 5G Reverse Transcriptases, murine RNase inhibitor, heat-labile UDG, and antibody-based hot start Taq DNA polymerase at optimal concentrations for consistent and efficient PCR amplification. For a 20 ul reaction, simply add 4 ul of 5x probe-based multiplex one-step qRT-PCR master mix to specific primers, DNA template, 50x ROX reference dye 1 and 2, and water. 

The probe-based multiplex one-step qRT-PCR kit is specially designed for single-plex or multiplex qPCR detection using RNA (e.g. SARS-COV-2 virus RNA) as template. It has extremely high stability, which can be stored at 37℃ for 14 days and its performance remains unchanged, and remains liquid form at a storage temperature of - 20℃ to avoid the thawing process of reagents. The reaction can be started by directly adding the template, primers and target probes required for detection. Using gene specific primers (GSP), the reverse transcription and qPCR can be finished in one tube, significantly reducing pipetting procedures and the risk of contamination. The probe-based multiplex one-step qRT-PCR kit contains a dUTP/UDG system that can prevent the contamination of PCR product. Heat-labile UDG can quickly eliminate the contamination of dU-containing products at room temperature. It can be inactivated at 55℃ without affecting the efficiency and sensitivity of qRT-PCR. Combining the superior performance of 5G Reverse Transcriptase and antibody-based hot-start Taq DNA Polymerase, with an optimized buffering system, the detection sensitivity of the probe-based multiplex one-step qRT-PCR kit can reach 0.1 pg of total RNA or less than 10 copies of RNA templates and is suitable for high-specificity detection systems based on fluorescence labelled probes (e.g. TaqMan).

Optimal template and primer concentrations, and qPCR program parameters should be determined experimentally by the investigator.

RNase-free H2O.

Storage/Stability: Stable for 1 year at -70°C. For maximum recovery of the product, centrifuge the original vial after thawing and before removing the cap. Aliquot to avoid repeated freezing and thawing.
