MB123: Heat-Labile UDG

Heat-labile UDG (Uracil DNA Glycosylase, UNG) is a recombinant uracil DNA glycosylase from marine bacterium BMTU 3346. Uracil DNA Glycosylase, a highly conserved DNA repair enzyme, catalyzes the excision of uracil from uracil-containing single- and double-stranded DNA. Like the other UDG enzyems such as the E.coli UDG, heat-labile UDG prevents mutagenesis due to misincorporation of uracil during DNA synthesis or spontaneous cytosine deamination in DNA strands. Because it is sensitive to high temperature and can be irreversibly inactivated above 50℃ more quickly than the other UDGs, heat-labile UDG is suitable for LAMP, PCR/qPCR, and RT-PCR/RT-qPCR. Another advantage of heat-labile UDG is that it is unnecessary to freeze PCR products immediately after amplification or to store them at -70°C because even the active enzyme does not degrade dU-PCR products at +2 to +8°C for at least several hours.

Unit definition: One unit of heat-labile UDG is defined as the amount of enzyme required to liberate 1 nmol uracil from dU-containing DNA in one hour at 37°C.

E.coli UDG (Uracil DNA Glycosylase, UNG) is a recombinant uracil DNA glycosylase from E.coli. Uracil DNA Glycosylase, a highly conserved DNA repair enzyme, catalyzes the excision of uracil from uracil-containing single- and double-stranded DNA. Like the other UDG enzyems such as the heat-labile UDG, E.coli UDG prevents mutagenesis due to misincorporation of uracil during DNA synthesis or spontaneous cytosine deamination in DNA strands. Because it can be irreversibly inactivated above 50℃, E.coli UDG is suitable for LAMP, PCR/qPCR, and RT-PCR/RT-qPCR. dU-containing ss and dsDNAs are substrates for UDG whereas RNAs and normal dT-containing DNAs are not.

Unit definition: One unit of E.coli UDG is defined as the amount of enzyme required to liberate 1 nmol uracil from dU-containing DNA in one hour at 37°C.
