Background

The rat anti-mouse PD-1 monoclonal antibody (clone No. RMP1-14, rat IgG2a kappa) reacts with the mouse PD-1 protein (CD279 or programmed death-1) encoded by the mouse pdcd1 gene, a member of the CD28 family of the Ig superfamily. PD-1 has two ligands, PD-L1 and PD-L2, both of which belong to the B7 family. It has been shown that in mouse models of melanoma, tumor growth can be transiently arrested via treatment with the anti-mouse PD 1 antibody and the anti-mouse PD L1 antibody which block the interaction between the PD-L1 protein and its receptor PD-1 protein. The RMP1-14 antibody blocks the binding of both the mouse PD-L1 protein and the mouse PD-L2 protein to the mouse PD-1 protein.

Our recombinant RMP1-14 antibodies have a part (variable regions) or complete amino acid sequences of the rat anti-mouse PD-1 antibody (hybridoma clone name or number: RMP1-14). The murinized anti-mouse PD-1 recombinant antibody has not only the constant regions of the mouse IgG antibodies but also the murinized variable regions.

Cooperative effects of programmed cell death-1 blockade and radiation in a model of a poorly immunogenic breast carcinoma.

We have previously shown that radiation therapy (RT) converts poorly immunogenic tumors resistant to antibodies (mAbs) against the checkpoint receptor CTLA-4 into susceptible ones. Importantly, RT induced the upregulation of PDL-1 and PDL-2 on the tumor cells and infiltrating myeloid cells in the TSA mouse breast cancer, suggesting that PD-1 ligands may limit rejection of the irradiated tumor by T cells. PD-1 blocking mAb RMP1-14 (anti-mouse PD-1 antibody) was given on day 15 and every 4 days thereafter, and mice were followed for tumor growth. In contrast, when RT was combined with anti-PD-1 mAb (RMP1-14, anti-mouse PD-1 mAb), all of the mice completely rejected tumors by day 25. Overall, data suggest that the IgG2a anti-PD-1 mAb RMP1-14 (anti-mouse PD-1/CD279 antibody) selectively depletes CD8+PD-1hi TILs, possibly by ADCC, while enhancing the RT-elicited priming and activation of tumor-specific CD8+ T cells.

Antimetastatic Effects of Blocking PD-1 and the Adenosine A2A Receptor.

Adenosine targeting is an attractive new approach to cancer treatment, but no clinical study has yet examined adenosine inhibition in oncology despite the safe clinical profile of adenosine A2A receptor inhibitors (A2ARi) in Parkinson disease. Metastasis is the main cause of cancer-related deaths worldwide, and therefore we have studied experimental and spontaneous mouse models of melanoma and breast cancer metastasis to demonstrate the efficacy and mechanism of a combination of A2ARi in combination with anti-PD-1 monoclonal antibody (mAb). This combination significantly reduces metastatic burden and prolongs the life of mice compared with either monotherapy alone. Importantly, the combination was only effective when the tumor expressed high levels of CD73, suggesting a tumor biomarker that at a minimum could be used to stratify patients that might receive this combination. Purified anti-mouse PD-1 mAb (RMP1-14, anti-mouse PD-1 antibody), anti-mouse CTLA-4 mAb (UC10-4F10), anti-mouse Tim3 (RMT3-23) and control Ig (2A3) were purchased from BioXCell (West Lebanon) and used in the schedule and dose as indicated.

Selinexor synergizes with anti-PD-1 antibody and inhibits tumor growth and metastasis in syngeneic mouse models of KRAS mutant colorectal cancer.

Colorectal cancer (CRC) is the second leading cause of cancer related death. In approximately half of all CRC cases, oncogenic KRAS mutations underlie poor prognosis and drug resistance. We performed preclinical studies to evaluate the effectiveness of selinexor and the anti-PD-1 antibody RMP1-14 (anti-mouse PD-1 antibody) in syngeneic mouse models of KRAS mutant CRC. Mice were allocated to four groups: vehicle control, selinexor (15 mg/kg, PO, once weekly), RMP1-14 (300 µg/mouse, IP, once weekly), or the combination of selinexor and RMP1-14 (anti-mouse PD-1 mAb). The combination of selinexor and RMP1-14 (anti-mouse PD-1/CD279 antibody) significantly inhibited tumor growth and metastasis with a TGI of 98.9% (p<0.0001).

Monoclonal antibodies (mAb) that block programmed death (PD)-1 or cytotoxic T lymphocyte antigen (CTLA-4) receptors have been associated with durable clinical responses against a variety of cancer types and hold great potential as novel cancer therapeutics. Recent evidence suggest that targeted blockade of multiple immunosuppressive pathways can induce synergistic antitumor responses. In this study, we investigated whether targeted blockade of CD73, an ectonucleotidase that catabolizes the hydrolysis of extracellular adenosine monophosphate (AMP) to adenosine, can enhance the antitumor activity of anti-CTLA-4 and anti-PD-1 mAbs against transplanted and chemically induced mouse tumors. Anti-CD73 mAb significantly enhanced the activity of both anti-CTLA-4 and anti-PD-1 mAbs against MC38-OVA (colon) and RM-1 (prostate) subcutaneous tumors, and established metastatic 4T1.2 breast cancer. The maternal pain threshold gradually increases during pregnancy, especially in late pregnancy. A series of mechanisms underlying pregnancy-induced analgesia have been reported. However, these mechanisms are still not completely clear, and the underlying molecular mechanisms need further investigation. We examined the relationship between the antinociceptive effect and the expression level of programmed cell death ligand-1 (PD-L1) during pregnancy and further observed the changes in pain thresholds and expression levels of cytokines in late-pregnant mice before and after blockade of PD-L1 or programmed cell death-1 (PD-1).
