Background
The rat anti-mouse PD-1 monoclonal antibody (clone No. RMP1-14, rat IgG2a kappa) reacts with the mouse PD-1 protein (CD279 or programmed death-1) encoded by the mouse pdcd1 gene, a member of the CD28 family of the Ig superfamily. PD-1 has two ligands, PD-L1 and PD-L2, both of which belong to the B7 family. It has been shown that in mouse models of melanoma, tumor growth can be transiently arrested via treatment with the anti-mouse PD 1 antibody and the anti-mouse PD L1 antibody which block the interaction between the PD-L1 protein and its receptor PD-1 protein. The RMP1-14 antibody blocks the binding of both the mouse PD-L1 protein and the mouse PD-L2 protein to the mouse PD-1 protein.
The rat anti-mouse PD-L1 monoclonal antibody 10F.9G2 (rat IgG2b kappa) reacts with the mouse PD-L1 protein (programmed death ligand-1, B7-H1 or CD274), a member of the B7 family of the Ig superfamily. PD-1 has two ligands, PD-L1 and PD-L2. It is under investigation how the recombinant anti-mouse PD-1 / PD-L1 bispecific antibodies (RMP1-14 / 10F.9G2 and 10F.9G2 / RMP1-14) behave different from the individual monoclonal antibodies and their combination.
Our recombinant anti-mouse PD-1 / PD-L1 bispecific antibodies have a part (variable regions) or complete amino acid sequences of the rat anti-mouse PD-1 antibody (hybridoma clone name or number: RMP1-14) and the rat anti-mouse PD-L1 antibody (hybridoma clone name or number: 10F.9G2).
The in vivo grade recombinant rat anti-mouse PD-1 / PD-L1 bispecific antibody (mouse IgG2c-LALAPG kappa) was produced in mammalian cells.
Immunogen: The original rat hybridoma (clone name: RMP1-14) was generated by immunizing rats with mouse PD-1-transfected BHK cells. The original rat hybridoma (clone name: 10F.9G2) was generated by immunizing rats with the mouse PD-L1 cDNA and mouse PD-L1 CHO transfectants.
Clone: RMP1-14.1, the same variable region sequences as the rat anti-mouse PD-1 monoclonal antibody (clone number: RMP1-14). 10F.9G2.1, the same variable region sequences as the rat anti-mouse PD-L1 monoclonal antibody (clone number: 10F.9G2).
Isotype: mouse IgG2c, kappa.
Applications: immunohistochemistry (IHC), Flow Cytometry (FC), and various in vitro and in vivo functional assays.
Formulation: 0.2 μM filtered solution of 1x PBS.
Purity: >95% by SDS-PAGE under reducing conditions.

Answer: Clones RMP1-14 and 10F.9G2 are the most well-known anti-mouse PD1 and PDL1 antibodies available in the market. They are commonly used to block PD-1 signaling in vivo in murine tumor models and other mouse models. These two clones both have extensive multi-year publication records supporting them. In addition, the mouse IgG2c antibody is produced in certain inbred strains such as C57BL/6, C57BL/10, SJL, and NOD, which does not express the mouse IgG2a antibody; the mouse IgG2a antibody is produced in other inbred strains such as BALB/c and Swiss Webster mice, which does not express the mouse IgG2c antibody. Because the C57BL/6 mouse strain is used in most of animal tumor model research, it is better to use the IgG2c antibodies rather than the IgG2a antibodies. The format with the Fc silenced, Fc silent, or Fcs with silenced effector function, such as LALAPG mutation, is the most popular for anti-mouse PD1 antibodies (clones RMP1-14 and 29F.1A12) and anti-mouse PD-L1 antibodies (clone 10F.9G2).
