Flow cytometric identification of CD8α+ dendritic cells using N418 antibody in tumor microenvironment
Zhang L, et al. Nat Commun. 2023 Mar 17;14(1):1490. doi: 10.1038/s41467-023-37009-2. PMID: 36922576
Tumor single-cell suspensions were stained with N418 antibody (BioLegend) to identify CD8α+ DCs for flow cytometry sorting. N418+ cells were gated based on CD11c co-expression in multi-parameter flow cytometry. Flow cytometry revealed increased N418+ DC frequencies in angiogenic tumors. Sorted N418+ populations were subjected to scRNA-seq post-flow analysis. Angiogenic markers were upregulated in N418+ DC subsets.

N418 staining for flow cytometry of CD8+ T cells in viral infection models

Infected tissue lymphocytes were labeled with N418 antibody conjugated to PE for flow cytometric analysis of CD8α expression. N418 staining combined with CD44 showed memory T cell subsets. Flow cytometry confirmed N418+ CD8 T cells in inflamed tissues. Activated CD8 subsets were quantified using N418-based gating. Data were acquired on a BD FACSCanto II flow cytometer.

Multi-parameter flow cytometry for N418+ CD8α DC subsets in spleen

Splenic cells were stained with N418 antibody for multi-color flow cytometry. N418 identified three distinct CD8α+ DC subsets in naive mice. Flow panels included N418 alongside CD103 and XCR1 markers. DC subset frequencies were quantified using N418 gating strategies. Cytokine profiles differed in N418+ populations.

Flow cytometric detection of N418+ cells in bone marrow-derived DCs

BMDCs were stained with N418 antibody to track CD8α expression via flow cytometry. N418+ DCs showed upregulated MHC II post-stimulation. Flow cytometry confirmed N418 on conventional DCs. Maturation markers were assessed in N418+ gates. FACS sorting isolated N418+ DCs for functional assays.

Characterization of thymic CD8α+ DCs by flow cytometry with N418 antibody

Thymic cells were stained with N418 antibody for flow cytometric analysis of CD8α+ DCs. N418+ DCs co-expressed DEC-205 in the thymus. Flow cytometry showed N418+ DCs supporting T cell selection. Progenitor interactions were quantified in N418-gated populations. Thymic DCs were analyzed using a BD LSR II flow cytometer.
