Source: The anti-mouse CD11a (LFA-1α) monoclonal antibody (clone: M17/4.4.11.9) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: M17/4.4.11.9) specifically binds to mouse CD11a (LFA-1α) .
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse CD11a (LFA-1α) protein.

Anti-CD11a ameliorates disease in the human psoriatic skin-SCID mouse transplant model: comparison of antibody to CD11a with Cyclosporin A and clobetasol propionate

After the healing period, animals transplanted with normal or psoriatic skin were treated for 14 days by daily intraperitoneal injection of either Cyclosporin A or a monoclonal antibody to human CD11a, or by topical application of clobetasol propionate. Anti-CD11a therapy significantly reduced epidermal thickness in psoriatic xenografts. In vivo anti-CD11a administration inhibited T cell infiltration into the epidermis. Psoriatic skin histology showed normalized keratinocyte differentiation post-anti-CD11a. Combination of anti-CD11a with cyclosporin A enhanced therapeutic effects.

Late treatment with anti-LFA-1 (CD11a) antibody prevents cerebral malaria in a mouse model

CBA/Ca mice injected with Plasmodium berghei were treated i.p. with anti-LFA-1 (CD11a) antibody (M17/4 clone) on days 4 to 6 after infection. Anti-LFA-1 treatment prevented the development of cerebral malaria symptoms. In vivo anti-CD11a blockade reduced brain sequestration of parasitized erythrocytes. Leukocyte accumulation in cerebral vessels was inhibited by anti-CD11a. Survival rates increased to 80% in the anti-CD11a-treated group.

Blockade of either CD28 or MHC class I inhibits experimental autoimmune encephalomyelitis

EAE-induced mice were administered anti-mouse CD11a antibody (M17/4.4.11.9 clone) i.v. to block CD11a in vivo. Anti-mouse CD11a monotherapy attenuated clinical EAE scores. In vivo CD11a blockade reduced CNS T cell infiltration. Demyelination lesions were minimized in anti-mouse CD11a-treated spinal cords. Combination with anti-CD28 further enhanced anti-mouse CD11a effects.

Tissue distribution and receptor-mediated clearance of anti-CD11a antibody in mice

Mice were injected i.v. with rat anti-mouse CD11a monoclonal antibody (M17/4 clone) to assess tissue distribution in vivo. Anti-mouse CD11a primarily distributed to leukocytes and macrophages in the spleen and liver. In vivo anti-CD11a uptake in lymph nodes increased after 72 h. Antibody was internalized by peripheral blood mononuclear cells in a time-dependent manner. LysoTracker staining confirmed lysosomal degradation of anti-mouse CD11a.

M17/4-mediated CD11a inhibition suppresses graft-versus-host disease in vivo

GVHD mice received M17/4.4.11.9 antibody i.p. to inhibit CD11a on donor T cells in vivo. M17/4 treatment reduced intestinal pathology and clinical GVHD scores. In vivo CD11a blockade decreased donor T cell proliferation in target organs. Skin and liver inflammation was attenuated post-M17/4 administration. Survival was prolonged in the M17/4 cohort compared to controls.
