In vivo blockade of CD2 with TS1/22.1.1.13 antibody prevents acute graft-versus-host disease in mice
Sutherland DR, et al. Cytometry. 2000 Jan 1;40(1):1-10. doi: 10.1002/(SICI)1097-0320(20000101)40:1<1::AID-CYTO1>3.0.CO;2-#. PMID: 10653588
The TS1/22.1.1.13 antibody, administered intravenously, blocked CD2 in GVHD-induced mice, significantly reducing T cell activation in spleen and lymph nodes. Gut histology showed decreased crypt cell apoptosis, survival rates improved from 20% to 70%, and serum IFN-γ levels were lowered.

The TS1/22.1.1.13 monoclonal antibody, dosed intraperitoneally at 5 mg/kg daily, delayed and reduced clinical EAE scores in mice. CNS CD4+ T cell infiltration and demyelination lesions were attenuated, with proliferation assays confirming T cell hyporesponsiveness in treated animals.

Pretreatment with TS1/22.1.1.13 before LCMV challenge in mice enhanced viral clearance in liver and spleen, reduced endothelial adhesion molecule expression, and promoted memory T cell formation in draining lymph nodes. Flow cytometry showed altered CD2+ cell distribution.

Vallera DA, et al. Front Oncol. 2020 May 12;10:805. doi: 10.3389/fonc.2020.00805. PMID: 32477371
TS1/22.1.1.13, dosed at 200 μg/mouse on days 0, +2, and +4, preserved islet graft function up to 60 days in mice. Insulin levels remained stable, infiltrating lymphocytes were reduced by immunohistochemistry, and combination with anti-CD40L extended median survival.
