A human/mouse cross-reactive anti-CD11b antibody (clone M1/70) was conjugated to a DFO chelator and radiolabeled with Zr-89. These data establish that [89Zr]anti-CD11b Ab immunoPET targets CD11b+ cells (TAMCs) with high specificity in a mouse model of GBM. Quantifying TAMCs using non-invasive immunoPET could facilitate patient stratification for TAMC-targeted treatments and monitoring of treatment efficacy. As TAMCs uniformly express the cell surface marker, integrin CD11b, we evaluated a Zr-89 labeled anti-CD11b antibody for non-invasive imaging of TAMCs in a syngeneic orthotopic mouse glioma model. CD11b is a novel alternate receptor for CD154 during alloimmunity

WT B6 recipients received BALB/c skin grafts and 106 DST, and either no further treatment, anti-CD40 alone, or anti-CD40 plus anti-CD11b (clone M1/70) on days 0, 2, 4, 6 as described in Materials and Methods. Blockade of CD154-CD11b interaction synergizes with anti-CD40 mAb to prolong graft survival. WT B6 recipients received BALB/c skin grafts plus 106 DST, and either no further treatment or anti-CD40 plus either the CD154/CD11b antagonist peptide cM7, or scM7 control peptide as described in Methods. MACS-enriched CD11b+ cells (purity 60%) from CD40?/? mouse splenocytes were incubated with 20 ?g /ml of mouse soluble CD154 or media alone as a control. Data indicate that the addition of anti-CD11b significantly improved allograft survival relative to blockade of CD40 alone (p<0.05).

CD11b+ neutrophils predominate over RAM11+ macrophages in stretch-injured muscle

The Mab.198 antibody was used to detect CD11b(+) neutrophils or macrophages. Neutrophils were identified specifically by using the RPN3 antibody. M1/70 attenuates blood-borne neutrophil oxidants, activation, and myofiber damage following stretch injury. The purpose of this study was to determine whether both neutrophils and macrophages infiltrate the hematoma site of stretch-injured rabbit tibialis anterior muscle. Toumi H, et al. Anti-CD11b monoclonal antibody improves myocardial function after six hours of hypothermic storage

Group M1/70 (n = 6) received at reperfusion 15 micrograms/mL of a monoclonal antibody F(ab')2 fragment to CD11b, the alpha-subunit of the leukocyte adhesion molecule Mac-1. Administration of M1/70 resulted in improved postischemic recovery of ventricular function compared with the two control groups (p < 0.05). These data implicate leukocyte-endothelial interactions mediated by the leukocyte adhesion molecule CD11b in myocardial dysfunction after long-term hypothermic ischemia. We studied a total of 20 hearts subjected to 6 hours of cardioplegic arrest at 4 degrees C. Specific antiadhesion strategies such as this may safely extend storage time for pediatric donor hearts.

Role of CD11b in tumor-associated macrophage polarization in hepatocellular carcinoma

Hepatocellular carcinoma-bearing mice were treated with anti-mouse CD11b antibody (M1/70 clone) to block CD11b on TAMs in vivo. Anti-mouse CD11b administration shifted TAM polarization from M2 to M1 phenotype. In vivo CD11b blockade reduced tumor growth and vascular density in HCC models. Tumor-associated macrophages were depleted in anti-mouse CD11b cohorts. Combination with sorafenib enhanced anti-mouse CD11b efficacy against HCC.
