Background:
The 1D3 antibody binds to the mouse CD19 protein, a transmembrane protein expressed in all B lineage cells including mouse plasma cells. CD19 plays two major roles in mouse B cells: it acts as an adaptor protein to recruit cytoplasmic signaling proteins to the membrane, and works within the CD19/CD21 complex to decrease the threshold for B cell receptor signaling pathways. CD19 is a biomarker for B lymphocyte development, lymphoma diagnosis and can be utilized as a target for leukemia immunotherapies.

Cells were stained with FITC-conjugated anti-CD19 mAb (clone 1D3) and analyzed by flow cytometry to monitor B cell depletion. The depletion efficiency was >95% in peripheral blood as determined by flow cytometry using 1D3 antibody. Peripheral blood was collected and stained with anti-CD19 (1D3) and anti-CD3 antibodies. Flow cytometry was performed on a FACSCalibur using CellQuest software. B cell depletion was confirmed by the absence of CD19+ cells in flow analysis.

For flow cytometry, cells were stained with anti-CD19 (clone 1D3) antibody to identify B cells. The percentage of CD19+ B cells was significantly reduced in treated mice. Spleen cells were isolated and stained with 1D3 antibody for CD19 expression. Data were acquired on a BD FACSCanto II flow cytometer. Treatment led to long-term B cell depletion as measured by flow.

B cells were identified by staining with PE-conjugated anti-CD19 (clone 1D3) antibody. Flow cytometry analysis showed eradication of CD19+ cells in lymphoma. The antibody 1D3 was used to gate B cell population in bone marrow. Peripheral blood B cells were monitored weekly using 1D3. CAR-T cells specifically targeted CD19+ cells as confirmed by flow.

Cells were stained with anti-CD19 mAb (1D3) for flow sorting. Peritoneal cells were analyzed for CD19 expression using 1D3. The antibody 1D3 was used in all flow cytometry experiments for B cell identification. Adoptive transfer experiments used sorted CD19+ cells with 1D3. B-1 cell self-renewal was assessed by limiting dilution analysis after sorting with 1D3.

Murine B cells were stained with biotinylated 1D3 anti-CD19 mAb followed by streptavidin-PE. Flow cytometry was used to assess CD19 expression levels during development. The 1D3 clone was specific for murine CD19. Developmental stages were gated based on 1D3 staining intensity. Signaling was correlated with CD19 surface density measured by 1D3.
