Background:

The B43 antibody binds to the human CD19 protein, a transmembrane protein expressed in all B lineage cells including human plasma cells. CD19 plays two major roles in human B cells: it acts as an adaptor protein to recruit cytoplasmic signaling proteins to the membrane, and works within the CD19/CD21 complex to decrease the threshold for B cell receptor signaling pathways. CD19 is a biomarker for B lymphocyte development, lymphoma diagnosis and can be utilized as a target for leukemia immunotherapies.

The light chain of the B43 antibody (anti-human CD19) was replaced in the pMPGB43k expression vector, which carries an enhanced CMV enhancer/promoter and a hygromycin resistance gene, to construct a new vector for recombinant protein expression. The B43 antibody (anti-human CD19 mAb) coding sequence was modified via PCR and cloned into the PmeI site of pMPGB34k, with a BamHI site added for further modifications. The study also isolated the IGF-1 coding sequence from murine cDNA for constructing pMPG-IGF-E5.

The B43 antibody (anti-human CD19) was used alongside W6/32 (MHC class I), RPA-T8 (CD8), M-A712 (transferrin receptor), and TU39 (MHC class II) for fluorescence-activated cell sorting and confocal immunofluorescence analyses. K3 and K5 proteins specifically downregulated MHC class I without affecting MHC class II, immunoglobulin M, or CD19 surface levels, as confirmed by these antibodies.

Large scale manufacturing of B43(anti-CD19)-genistein for clinical trials in leukemia and lymphoma

The B43(anti-human CD19 antibody)-genistein immunoconjugate was manufactured at large scale for clinical trials in leukemia and lymphoma. The B43 antibody was purified from ascites fluid using protein A affinity chromatography, and its conjugation to genistein was optimized for high yield. Flow cytometry confirmed effective binding of B43-genistein to CD19+ leukemic cells, with cytotoxicity tested against leukemia cell lines.

Pharmacokinetic features, immunogenicity, and toxicity of B43(anti-CD19)-pokeweed antiviral protein immunotoxin in cynomolgus monkeys

The B43(anti-human CD19)-pokeweed antiviral protein (PAP) immunotoxin was evaluated in cynomolgus monkeys for pharmacokinetics, immunogenicity, and toxicity. B43-PAP, constructed by linking the anti-CD19 monoclonal antibody B43 to PAP, killed >99.9% of primary leukemic progenitor cells from B-lineage ALL patients. The study revealed dose-limiting renal and cardiac toxicity in mice and severe renal tubular necrosis in monkeys.

The B43 monoclonal antibody (anti-human CD19 mAb) was used to cross-link membrane-associated CD19, activating the LYN kinase signaling pathway and inducing apoptosis in B-lineage lymphoma cells in a p53- and Bcl-2-independent manner. The CD19-LYN complex was immunoprecipitated using B43 antibody, and flow cytometry confirmed high-level CD19 expression on lymphoma cells.
