Source: The anti-human CD19 monoclonal antibody (clone: FMC63) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: FMC63) specifically binds to the human CD19 protein.
Applications: ELISA, flow cytometry, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human CD19 protein.

Background

The FMC63 antibody binds to the human CD19 protein, a transmembrane protein expressed in all B lineage cells including human plasma cells. CD19 plays two major roles in human B cells: it acts as an adaptor protein to recruit cytoplasmic signaling proteins to the membrane, and works within the CD19/CD21 complex to decrease the threshold for B cell receptor signaling pathways. CD19 is a biomarker for B lymphocyte development, lymphoma diagnosis and can be utilized as a target for leukemia immunotherapies.

The FMC63 antibody (anti-human CD19) is the basis for four of six FDA-approved CAR T cell therapies targeting CD19 for B-cell malignancies. Despite its widespread use, reported affinities of FMC63-scFv to CD19 vary from 0.3 to 47 nM. This study conducted a detailed affinity analysis of the FMC63-CD19 interaction to resolve these inconsistencies, highlighting its critical role in CAR T cell therapy development.

The FMC63 scFv (anti-human CD19) is used in all four commercial CAR T cell products, targeting a membrane-distal CD19 epitope with slow on- and off-rates. A novel anti-CD19 scFv (h1218) targeting a membrane-proximal epitope with faster kinetics outperformed FMC63-CAR T cells in eradicating lymphomas with CD19 mutations (L174V, R163L) in vivo, showing reduced activation-induced cell death and enhanced expansion.

Selective B cell depletion upon intravenous infusion of replication-incompetent anti-CD19 CAR lentivirus

A second-generation anti-CD19 CAR with FMC63 scFv (anti-human CD19) showed robust B-cell targeting in vitro, with 18.5% ± 1.08% of murine B cells unviable after 5-hour co-culture. Compared to 1D3 (anti-mouse CD19), FMC63-CAR T cells achieved an 8.8-fold expansion in transduced splenocytes, demonstrating selective and effective B-cell depletion in vivo using a standard CAR design.

Fully human CD19-specific chimeric antigen receptors for T-cell therapy

Human CD19-specific scFvs with binding characteristics similar to FMC63 (anti-human CD19) were generated from human Ab/DNA-libraries, targeting an overlapping CD19 epitope. These scFvs were incorporated into CARs and compared to FMC63-based CARs, which are used in clinical trials. Flow cytometry with FMC63 competition confirmed their binding specificity, demonstrating comparable T-cell redirection for B-cell malignancy treatment.
