Background
The 1D3 antibody binds to the mouse CD19 protein, a transmembrane protein expressed in all B lineage cells including mouse plasma cells. CD19 plays two major roles in mouse B cells: it acts as an adaptor protein to recruit cytoplasmic signaling proteins to the membrane, and works within the CD19/CD21 complex to decrease the threshold for B cell receptor signaling pathways. CD19 is a biomarker for B lymphocyte development, lymphoma diagnosis and can be utilized as a target for leukemia immunotherapies.

Selective B cell depletion upon intravenous infusion of replication-incompetent anti-CD19 CAR lentivirus.

To explore this approach we administered approximately 20 million replication-incompetent VSV-G lentiviral particles carrying an anti-CD19CAR-2A-GFP transgene comprising either an FMC63 (human) or 1D3 (murine) anti-CD19 binding domain, or a GFP-only control transgene, to wild-type C57BL/6 mice by tail vein infusion. We constructed a CAR where we changed the scFv binding domain from FMC63[19] (a monoclonal antibody raised against human CD19) to 1D3[20] (a monoclonal antibody raised against mouse CD19). Specifically, we saw emergence of a persistent 1D3-CD19CAR-GFP transduced CD3 + T cell population beginning at week 3 that reaching a maximum of 13.5 +/- 0.58% (mean +/- SD) of the peripheral blood CD3 + T cell population by week 5 [Figure 2A-2C]. The only overt change to mice treated with either 1D3- or FMC63-CD19CAR lentivirus was a modest decrease in weight of 5.5 +/- 2.97% (mean +/- SD) and 3.6 +/- 1.78%, respectively over the first week after treatment [Figure S6]. We conducted qPCR (Taqman) analysis to test for the presence of the CD19CAR transgene in additional tissues obtained 2 or 6 weeks after infusing mice with either of two different doses (2x10^7 or 2x10^6 IU) of the 1D3-CD19CAR-GFP virus.

For B cell depletion, a combination of 300 µg of anti-mouse CD19 antibody (clone 1D3) and 300 µg of anti-mouse B220 antibody (clone RA3.3A1/6.1) was intraperitoneally injected 3 weeks before tumor inoculation and every 5 days thereafter [25]. To establish in vivo models, 5×10^6 luciferase-tagged ID8 (ID8-luc) cells were intraperitoneally injected into Six-week-old C57BL/6 mice. An in vivo anti-mouse PD-1 antibody (clone RMP1-14) was purchased from... Three weeks after cell implantation, mice were peritoneally injected with abemaciclib (50 mg/kg in 200 μL), an anti-PD-1 mAb (200 μg in 100 μL per injection), abemaciclib plus the anti-PD-1 mAb, or PBS (200 μL per injection). For in vivo neutralization of CXCL10, 50 μg of Rat anti-mouse CXCL10 was intraperitoneally injected into tumor bearing mice every 3 days for 3 weeks, Rat IgG was used as isotype control.

Stimulation with FITC-labeled antigens confers B cells with regulatory properties.

After incubation, cells were washed and stained with a PE-labeled anti-mouse CD19 antibody (1D3) for 25 min at 4°C. Endocytosis of antigens was performed using negatively selected B cells purified as described above. B cells were incubated alone or in the presence of antigens for 1 hr at 4°C or 37°C. After incubation, B cells were washed, stained with a PE-labeled anti-mouse CD19 antibody (1D3) for 25 min at 4°C, and permeabilized with the Cytofix/Cytoperm kit. When indicated, the FITC and Pacific blue fluorescences were normalized using FITC-labeled and Pacific Blue-labeled anti-CD19 IgG (clone 1D3). B-cell activation was evaluated using Pacific blue-conjugated anti-CD19 (1D3), PE-conjugated anti-CD80, APC-conjugated anti-CD83 (Michel-19), PE-conjugated anti-I-A b /I-E b (2G9) and biotinylated anti-CD86 (GL1) associated with the Alexa-fluor® 700 labeled streptavidin (Molecular probes®, Invitrogen).

Anti-CD19 Ab (clone 1D3), LY294002, and BP-1-102 were also used for the following experiments. The therapeutic agents, including paclitaxel, cisplatin, bevacizumab, and olaparib, diluted with DMSO, was intraperitoneally administered to mice. Anti-BTLA Ab (clone 6A6) , anti-PD-1 Ab (clone RMP1–14) , anti-PD-L1 Ab (clone 10F.9G2), anti-CD19 Ab (clone 1D3), LY294002, and BP-1-102 were also used for the following experiments. Briefly, C57BL/6 J mice (10 mice per group) were intraperitoneally challenged with 1 × 10^5 WF-3/Luc tumor cells on day 0. Mice treated with paclitaxel 6 mg/kg and anti-CD19 Ab 30 μg/mouse lived longer than those treated with paclitaxel or anti-CD19 Ab alone (p = 0.004, log-rank test, Additional file 5: Figure S5).

The antibodies used for mouse staining were anti-CD19 (1D3), anti-CD31 (390), anti-CD45 (30-F11), anti-TER119 (TER-119), and anti-CD140a (APA5). For in vivo experiments, mice were injected with 10^6 CAR T cells intravenously. The brain was harvested and processed for flow cytometry. CAR T cell infiltration was assessed using the antibodies listed. No 1D3-specific sentence, but the antibody is used for staining in the methods.
