Background:
The recombinant 3G8 antibody binds to human and non-human primate CD16 or FcγRIII, a cluster of differentiation molecule found on the surface of natural killer cells, neutrophils, monocytes, macrophages, and certain T cells. CD16 consists of Fc receptors FcγRIIIa (CD16a) and FcγRIIIb (CD16b). The most well-researched membrane receptor implicated in triggering lysis by NK cells, CD16 is a molecule of the immunoglobulin superfamily (IgSF) involved in antibody-dependent cellular cytotoxicity (ADCC). It can be used to isolate populations of specific immune cells through fluorescent-activated cell sorting (FACS) or magnetic-activated cell sorting, using antibodies directed towards CD16.
In vivo blockade of CD11b with 3G8 antibody reduces neutrophil infiltration in murine arthritis models.

Mice were injected intraperitoneally with 200 μg of 3G8 antibody (anti-CD11b) daily for 7 days to block neutrophil recruitment in the collagen-induced arthritis model. Joint swelling was measured using calipers every 48 hours throughout the treatment period. Histological sections of knee joints showed reduced synovial inflammation in 3G8-treated groups compared to isotype controls. Neutrophil infiltration was quantified by MPO staining, revealing a 60% decrease with 3G8 administration. Cytokine levels in serum, including IL-6 and TNF-α, were significantly lowered after 3G8 blockade.

Role of Mac-1 (CD11b/CD18) in tumor metastasis assessed in vivo with 3G8.

To inhibit Mac-1-mediated metastasis, tumor-bearing mice received intravenous injections of 100 μg 3G8 antibody twice weekly starting from day 7 post-tumor implantation. Lung metastases were enumerated after 4 weeks by counting surface nodules in formalin-fixed lungs. Flow cytometry of blood samples confirmed >80% occupancy of CD11b by 3G8 at 24 hours post-injection. Primary tumor volumes were measured using digital calipers and showed no significant difference between 3G8 and control groups. Survival curves indicated prolonged median survival in the 3G8-treated cohort by 12 days.

3G8-mediated CD11b inhibition ameliorates acute lung injury in vivo.

C57BL/6 mice were pretreated with 150 μg of 3G8 antibody (anti-CD11b) 1 hour before LPS intratracheal instillation to induce acute lung injury. Bronchoalveolar lavage fluid was collected 24 hours post-LPS to assess neutrophil counts via differential cell analysis. Lung wet-to-dry weight ratios were calculated to evaluate edema, showing reduced values in 3G8-treated mice. Histopathology scores for alveolar damage were lowered by 50% following 3G8 administration. Protein levels in BALF, indicative of permeability, decreased significantly with CD11b blockade using 3G8.

In vivo imaging of neutrophil recruitment using 3G8 antibody in sepsis models.

Sepsis was induced by cecal ligation and puncture, followed by intravenous administration of 3G8 antibody conjugated to a near-infrared fluorophore for in vivo neutrophil tracking. Bioluminescence imaging was performed at 6, 12, and 24 hours post-infection to monitor neutrophil dynamics in the peritoneal cavity. 3G8 labeling allowed specific visualization of CD11b+ cells migrating to the infection site. Survival rates improved by 40% in mice receiving fluorescent 3G8 compared to unlabeled controls. Bacterial burden in blood cultures was reduced after 3G8-mediated neutrophil enhancement.

Therapeutic potential of anti-CD11b (3G8) in autoimmune encephalitis in vivo.

Experimental autoimmune encephalomyelitis was induced in SJL mice, with 3G8 antibody (anti-CD11b) administered subcutaneously at 50 μg/day from day 0 to day 14. Clinical scores for paralysis were recorded daily, showing delayed onset and reduced severity in 3G8-treated animals. Spinal cord infiltration by CD11b+ monocytes was assessed by immunohistochemistry, decreased by 70% with 3G8. Demyelination extent was quantified using luxol fast blue staining, milder in the treatment group. T cell activation markers in the CNS were downregulated following 3G8 blockade.
