Source: The anti-mouse/human VLA-4 (CD49d) monoclonal antibody (clone: PS/2) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: PS/2) specifically binds to mouse/human VLA-4 (CD49d).
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse/human VLA-4 (CD49d) protein.

Anti-VLA-4 antibody treatment reduces inflammation in a mouse model of colitis

The PS/2 antibody, specific for mouse VLA-4 (CD49d), was administered intraperitoneally at 100 ?g every other day for 7 days to block leukocyte adhesion in a mouse colitis model. Colonic inflammation was assessed by histological scoring, and cytokine levels (IL-6 and TNF-?) were measured by ELISA. VLA-4 blockade with PS/2 reduced inflammatory cell infiltration by 50%, demonstrating its anti-inflammatory efficacy.

In vivo blockade of VLA-4 with PS/2 antibody prevents EAE progression in mice

The anti-mouse VLA-4 (PS/2) antibody was injected intravenously to inhibit T cell migration in a mouse model of experimental autoimmune encephalomyelitis (EAE) induced by MOG35-55 peptide in CFA. Clinical scores were monitored daily for 21 days, and spinal cord infiltrates were analyzed by flow cytometry. PS/2 treatment reduced disease severity by 60% compared to controls, highlighting its role in preventing EAE progression.

PS/2 monoclonal antibody inhibits leukocyte adhesion in vivo during peritonitis

The PS/2 monoclonal antibody was administered intraperitoneally at 200 ?g to block VLA-4-mediated leukocyte adhesion in a thioglycollate-induced peritonitis model in mice. Leukocyte recruitment was quantified in peritoneal lavage fluid, showing a 70% reduction in neutrophil infiltration in PS/2-treated mice. VCAM-1 expression was confirmed by immunohistochemistry, supporting the role of PS/2 in inhibiting adhesion.

In vivo administration of PS/2 antibody blocks T cell migration to inflamed sites

The PS/2 monoclonal antibody, targeting VLA-4, was administered at 100 ?g per dose to block T cell migration to inflamed tissues in a mouse contact hypersensitivity model. T cell infiltration was assessed in skin biopsies, and ear swelling was significantly reduced in PS/2-treated mice, demonstrating the antibody’s efficacy in inhibiting T cell migration to inflamed sites.

In vivo role of VLA-4 in neutrophil recruitment during acute lung injury
