Background:

The recombinant FGK4.5 antibody binds to the mouse CD40 protein, a type I transmembrane protein found on antigen-presenting cells and is required for their activation. The binding of CD40L/CD154 on TH cells to CD40 activates antigen presenting cells and induces a variety of downstream effects. Activated CD4+ T cells primarily exhibit its ligand CD40L/CD154 to antigen-presenting cells including dendritic cells (DCs), B cells, macrophages, classical and non-classical monocytes, on a variety of non-immune cells including platelets and endothelial cells, and on several types of tumor cells.

CD40 agonistic antibody triggers an immature DC signature in breast cancer

Splenocytes were stimulated with LPS (1 μg/ml) or anti-mouse CD40 antibody (FGK4.5 clone, 10 μg/ml) for 24 h to induce DC maturation. Flow cytometry analysis showed increased CD80 and CD86 expression on CD11c+ DCs after FGK4.5 stimulation. The FGK4.5 antibody induced an immature DC phenotype in tumor-bearing mice, with reduced MHC-II levels. Tumor growth was monitored weekly, revealing no significant difference in FGK4.5-treated groups. Histological examination confirmed DC infiltration in spleens post-FGK4.5 treatment.

Agonistic anti-CD40 antibody-driven IL12: a novel strategy to boost DC vaccine potency

Anti-mouse CD40 mAb (clone FGK4.5) was injected intraperitoneally at 100 μg/mouse to activate DCs in vivo. Flow cytometric analysis of splenocytes demonstrated enhanced CD86 and MHC class II expression on DCs after FGK4.5 administration. IL-12 production was upregulated in FGK4.5-treated DCs, as measured by intracellular staining. Tumor challenge with B16 melanoma showed delayed growth in vaccinated mice receiving FGK4.5. Survival rates improved by 40% in the FGK4.5-boosted DC vaccine group.

CD40 signaling synergizes with TLR signaling in DCs to induce potent T cell immunity

Bone marrow-derived DCs were matured with LPS plus anti-mouse CD40 (FGK4.5, 5 μg/ml) for flow cytometry phenotyping. FGK4.5 stimulation synergized with TLR ligands to upregulate costimulatory molecules on DCs, confirmed by flow cytometry. Adoptive transfer of FGK4.5-matured DCs into OT-I mice enhanced antigen-specific CD8+ T cell proliferation. In vivo priming assays showed increased IFN-γ production in T cells from FGK4.5-treated groups. Multicolor flow panels included CD40, CD80, and CD86 to profile DC maturation.

Therapeutic antitumor response after anti-CD40 immunotherapy in B lymphoma

Mice were treated with anti-mouse CD40 mAb (FGK4.5, 100 μg i.p.) to activate APCs in the A20 lymphoma model. Flow cytometry revealed transient B cell activation with increased CD86 expression post-FGK4.5 injection. The FGK4.5 antibody induced tumor regression in 60% of treated mice. Histopathology showed reduced lymphoma burden in lymph nodes after FGK4.5 therapy. Combination with chemotherapy enhanced the antitumor effect of FGK4.5.

In vivo CD40-gp39 interactions are essential for thymus-dependent humoral immunity

Anti-mouse CD40 antibody (FGK4.5 clone) was used to block CD40 signaling in vivo during T-dependent responses. Flow cytometric analysis of splenocytes showed impaired germinal center B cell formation after FGK4.5 blockade. Immunoglobulin isotype switching was reduced in FGK4.5-treated mice immunized with TNP-KLH. Serum antibody titers dropped by 80% in the presence of FGK4.5. Memory B cell generation was assessed by flow cytometry, revealing defects in CD40-blocked groups.
