CD45RB antibody induces long-term immunological tolerance by regulating the function of B cells in a mouse model of transplantation

Recipient mice were treated with anti-mouse CD45RB antibody (MB23G2 clone) i.p. to induce tolerance in vivo. Anti-mouse CD45RB administration prolonged allograft survival in 80% of recipients. In vivo anti-mouse CD45RB blockade regulated B cell activation in the spleen. Humoral responses were suppressed post-anti-mouse CD45RB treatment. Long-term tolerance was achieved without chronic rejection in anti-mouse CD45RB cohorts.

Anti-CD45RB monoclonal antibody induces tolerance and promotes the generation of regulatory T cells in vivo

Skin graft recipients received anti-mouse CD45RB (MB23G2) antibody for tolerance induction in vivo. Anti-mouse CD45RB treatment increased Foxp3+ Treg frequencies in vivo. In vivo anti-mouse CD45RB promoted T cell anergy in allografts. Graft histology showed reduced infiltration post-anti-mouse CD45RB. Tolerance was transferable via anti-mouse CD45RB-treated splenocytes.

In vivo blockade of CTLA-4 enhances the priming of responsive T cells but fails to prevent the induction of tumor antigen-specific tolerance

Purified T cells were stained with Cy-Chrome-labeled anti-mouse CD4, biotinylated anti-TCR clonotype mAb 6.5 followed by phycoerythrin-labeled streptavidin and FITC-conjugated anti-mouse CD45RB. Forward light scatter (FSC) and the level of CD45RB expression were determined on clonotype+ T cells. The results are representative of two experiments with three mice per group. BALB/c mice received anti-HA TCR transgenic T cells on day 0 and were treated with the anti-CTLA-4 antibody 9H10 (100 μg/day, i.p.) on days −1, 0, and +1 or received no treatment. On day +2 after T cell transfer half the mice in each group were immunized with vacc-HA, and 6 days later, T cells were isolated as described in Materials and Methods.

Anti-CD45RB induces tolerance through preferential depletion of recently activated CD4+ T cells

Transplant recipients were administered anti-mouse CD45RB (MB23G2 clone) i.v. to deplete activated T cells in vivo. Anti-mouse CD45RB preferentially eliminated CD45RBlo activated CD4+ T cells. In vivo anti-mouse CD45RB treatment preserved naive T cell populations. Allograft rejection was prevented in anti-mouse CD45RB cohorts. Treg expansion was observed post-anti-mouse CD45RB administration.

CD45RB expression on CD4+ T cells correlates with the ability to induce or suppress graft-versus-host disease in vivo

GVHD mice received sorted CD4+ T cells with high or low CD45RB expression, treated with anti-mouse CD45RB (MB23G2) in vivo. Anti-mouse CD45RB depleted CD45RBhi effector T cells selectively. In vivo anti-mouse CD45RB shifted the balance toward suppression. GVHD scores decreased in anti-mouse CD45RB-treated recipients. CD45RBlo cells mediated tolerance post-anti-mouse CD45RB.
