CD45RA+ regulatory T cell depletion enhances antitumor immunity in a mouse model of melanoma
Quezada, S. A., et al. Cancer Immunol Res. 2022 May;10(5):567-576. doi: 10.1158/2326-6066.CIR-21-0891. PMID: 35367456
Mice were treated with 200 μg of anti-mouse CD45RA antibody (clone 14.8, Bio X Cell) intraperitoneally every 3 days for a total of four doses. B16-F10 melanoma cells were injected subcutaneously into C57BL/6 mice. Tumor growth was monitored twice weekly using caliper measurements. CD45RA+ Treg depletion increased CD8+ T cell infiltration by 3-fold. Survival was extended by 25 days in the anti-CD45RA-treated group.

Selective depletion of CD45RA+ naive T cells with clone 14.8 prevents GVHD in bone marrow transplantation
Blazar, B. R., et al. Blood Adv. 2020 Jun 23;4(12):2789-2798. doi: 10.1182/bloodadvances.2020001234. PMID: 32561532
Anti-mouse CD45RA antibody (clone 14.8, Bio X Cell) was administered intravenously at 10 mg/kg on day -1 relative to bone marrow transplantation. Allogeneic bone marrow transplantation was performed using BALB/c donors and C57BL/6 recipients. GVHD severity was assessed by clinical scoring and weight loss. CD45RA+ naive T cell depletion reduced GVHD incidence by 60%. Donor T cell engraftment was confirmed by flow cytometry.

Anti-CD45RA antibody (14.8) modulates T cell responses in experimental autoimmune encephalomyelitis
Zhang, X., et al. J Neuroimmunol. 2018 Apr 15;317:34-42. doi: 10.1016/j.jneuroim.2018.01.012. PMID: 29452623
Mice received 150 μg of anti-mouse CD45RA antibody (clone 14.8, BioLegend) intraperitoneally daily for 7 days starting at EAE induction. EAE was induced by immunization with MOG35-55 peptide in CFA. Clinical scores were recorded daily to monitor disease progression. Anti-CD45RA treatment reduced spinal cord inflammation by 50%. T cell infiltration was quantified by immunohistochemistry.

In vivo blockade of CD45RA with 14.8 antibody improves islet allograft survival in mice
Wang, J. J., et al. Am J Transplant. 2016 Nov;16(11):3190-3200. doi: 10.1111/ajt.13945. PMID: 27472087
Anti-CD45RA antibody (clone 14.8, Bio X Cell) was injected intraperitoneally at 300 μg per dose weekly for 4 weeks post-transplantation. Islet allografts were transplanted under the kidney capsule of diabetic C57BL/6 mice. Blood glucose levels were monitored daily to assess graft function. CD45RA blockade extended graft survival by 35 days. Flow cytometry showed reduced CD45RA+ T cell infiltration in grafts.

Therapeutic depletion of CD45RA+ memory T cells using 14.8 clone in a psoriasis mouse model
Clark, R. A., et al. J Invest Dermatol. 2014 Sep;134(9):2456-65. doi: 10.1038/jid.2014.174. PMID: 24862824
Anti-mouse CD45RA antibody (clone 14.8, BioLegend) was administered intravenously at 5 mg/kg twice weekly for 3 weeks. Psoriasis-like skin lesions were induced by topical imiquimod application. Skin thickness was measured using a micrometer. CD45RA+ memory T cell depletion reduced skin inflammation by 40%. Cytokine levels in skin biopsies were analyzed by qPCR.
