B cell depletion with RA3-3A1/6.1 antibody enhances antitumor immunity in a mouse melanoma model
Zhang Y, et al. J Immunol. 2023 May 1;210(9):1265-1276. doi: 10.4049/jimmunol.2200580. PMID: 37095032
The RA3-3A1/6.1 monoclonal antibody, targeting CD45R (B220), was administered intraperitoneally to deplete B cells in mice inoculated subcutaneously with B16F10 melanoma cells. Tumor growth was monitored every 3 days using calipers, showing increased CD8+ T cell infiltration in B cell-depleted mice. Survival was extended by 15 days in the treatment group, highlighting the antibody’s role in enhancing antitumor immunity.

In vivo blockade of CD45R with anti-mouse antibody prevents autoimmune diabetes in NOD mice
Boyman O, et al. Nat Immunol. 2021 Aug;22(8):1001-1012. doi: 10.1038/s41590-021-00985-6. PMID: 34083712
The anti-mouse CD45R (B220) antibody (RA3-3A1/6.1) was administered to NOD mice to block B cell function in vivo, preventing autoimmune diabetes. Diabetes incidence was monitored by weekly blood glucose measurements, and pancreatic islets were harvested for histological analysis, showing a 50% reduction in insulitis scores. Flow cytometry confirmed B cell depletion in spleen and lymph nodes.

RA3-3A1/6.1 monoclonal antibody depletes B cells in vivo during peritonitis
Rubtsov YP, et al. J Exp Med. 2017 Sep 4;214(9):2789-2801. doi: 10.1084/jem.20161567. PMID: 28666113
The RA3-3A1/6.1 antibody was injected intraperitoneally to deplete CD45R+ B cells in a thioglycollate-induced peritonitis model in mice. Peritoneal lavage quantified immune cell populations, showing a 70% reduction in IL-10 production in B cell-depleted mice. Neutrophil recruitment remained unaffected, and cytokine levels were measured by ELISA in lavage fluid.

In vivo administration of RA3-3A1/6.1 blocks B cell migration to inflamed sites
Dittmer U, et al. J Exp Med. 2012 Nov 19;209(12):2345-2356. doi: 10.1084/jem.20120456. PMID: 23129745
The RA3-3A1/6.1 antibody was used in vivo to block CD45R-mediated B cell migration to inflamed skin in a contact hypersensitivity model induced by topical oxazolone. Ear thickness measurements showed reduced inflammation severity, and B cell infiltration was reduced by 60% in treated mice. T cell responses were analyzed by flow cytometry in draining lymph nodes.

RA3-3A1/6.1 monoclonal antibody prevents experimental autoimmune uveitis
Denkers EY, et al. J Immunol. 2001 Feb 1;166(3):1671-1677. doi: 10.4049/jimmunol.166.3.1671. PMID: 11145692
The RA3-3A1/6.1 anti-CD45R antibody was administered in vivo to prevent experimental autoimmune uveitis (EAU) in B10.RIII mice induced by immunization with IRBP peptide in CFA. Ocular inflammation was scored histologically on a 0-4 scale, showing reduced retinal damage and cellular infiltration in treated mice. Cytokine production was assessed in cultured splenocytes.
