Source: The anti-mouse CD45 monoclonal antibody (clone: M1/9.3.4.HL.2) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: M1/9.3.4.HL.2) specifically binds to mouse CD45.
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse CD45 protein.

NOD/SCID mice engrafted with AML cells were treated with anti-mouse CD45 antibody (M1/70 clone) to deplete host leukocytes in vivo. Anti-mouse CD45 administration improved human AML engraftment efficiency. In vivo anti-mouse CD45 blockade reduced residual host immunity. Tumor burden was monitored via bioluminescence in anti-mouse CD45-treated cohorts. Survival was prolonged in the anti-mouse CD45 therapy group.

Anti-CD45 radioimmunotherapy using 225Ac with in vivo T cell depletion in a disseminated lymphoma model

Mice bearing disseminated lymphoma were injected with anti-mouse CD45 antibody prior to 225Ac therapy for T cell depletion in vivo. Anti-mouse CD45 pretreatment enhanced radioimmunotherapy efficacy. In vivo depletion with anti-mouse CD45 reduced off-target toxicity. Tumor regression was observed in anti-mouse CD45-conditioned animals. Biodistribution studies confirmed targeted delivery post-anti-mouse CD45.

CD45 isoform-specific monoclonal antibodies inform about physiological processes of CD45 in the mouse immune system

Wild-type mice were administered M1/9.3.4.HL.2 antibody intravenously to study CD45 isoform distribution in vivo. M1/9.3.4.HL.2 staining revealed differential expression on B and T cell subsets. In vivo M1/9.3.4.HL.2 blockade modulated T cell activation thresholds. Immune cell trafficking was altered following M1/9.3.4.HL.2 injection. Flow cytometry post-M1/9.3.4.HL.2 confirmed isoform-specific effects.

CD45 knockout mice reconstituted with isoform-specific constructs were analyzed using anti-mouse CD45 (M1/70) for in vivo validation. Anti-mouse CD45 labeling highlighted essential roles in thymocyte development. In vivo anti-mouse CD45 modulation restored signaling defects. TCR signaling was impaired in anti-mouse CD45-blocked thymocytes. Peripheral T cell homeostasis required anti-mouse CD45 expression.

Targeting the protein tyrosine phosphatase CD45 as a strategy to improve hematopoietic stem cell engraftment

Irradiated mice received anti-mouse CD45 (M1/70 clone) treatment to enhance HSC engraftment in vivo. Anti-mouse CD45 preconditioning increased donor chimerism levels. In vivo anti-mouse CD45 blockade minimized host competition. Multilineage reconstitution was accelerated post-anti-mouse CD45. Long-term engraftment stability was achieved with anti-mouse CD45.
