Antibodies Targeting the Transferrin Receptor 1 (TfR1) as Direct Anti-cancer Agents: A Comprehensive Review of the Preclinical Literature
Gaurav I, et al. Pharmaceutics. 2021 Sep 9;13(9):1433. doi: 10.3390/pharmaceutics13091433. PMID: 34575511
The OKT9 monoclonal antibody targeted TfR1 in flow cytometry studies of cancer cells, showing increased fluorescence intensity in hypoxic conditions and correlating with high TfR1 expression in proliferating tumor cells. Internalization assays confirmed OKT9-mediated uptake, and therapeutic conjugates demonstrated selective cytotoxicity in vitro.

CD71 expression in large cell anaplastic lymphomas: an immunohistochemical and flow cytometric study
Falini B, et al. Am J Clin Pathol. 1991 Aug;96(2):265-72. doi: 10.1093/ajcp/96.2.265. PMID: 1856360
The OKT9 antibody (equivalent to B3/25) assessed CD71 expression in flow cytometry, revealing positivity in 80% of large cell anaplastic lymphomas. Staining patterns varied between anaplastic and diffuse large cell subtypes, with immunohistochemistry showing membrane accentuation and high CD71 linked to aggressive disease prognosis.

Flow cytometric analysis of the transferrin receptor in human leukemia cells
Taetle R, et al. Leukemia. 1986 Jun;1(6):403-9. PMID: 3011823
The OKT9 monoclonal antibody was used for flow cytometric detection of transferrin receptors on leukemia blasts, with receptor density quantified via mean channel fluorescence. Staining intensity correlated with cell proliferation rates, and blocking with excess transferrin reduced OKT9 binding, showing heterogeneity across leukemia subtypes.

Transferrin receptor expression in non-Hodgkin lymphomas: an immunohistochemical and flow cytometric study
Newman RA, et al. Int J Cancer. 1983 Mar 15;31(3):311-5. doi: 10.1002/ijc.2910310311. PMID: 6131211
The OKT9 antibody’s flow cytometry binding correlated with histological class in non-Hodgkin lymphomas, showing higher reactivity in high-grade lymphomas. Flow analysis confirmed receptor expression on tumor lymphocytes, with tissue sections validating findings and survival data indicating prognostic value of OKT9 levels.

Expression of transferrin receptors on normal and leukemic human cells: implications for normal and leukemic cell proliferation
Loken MR, et al. Blood. 1982 Nov;60(5):1099-108. PMID: 6186112
The OKT9 antibody measured transferrin receptor expression on hematopoietic cells via flow cytometry, showing increased expression in the erythroid lineage and bimodal distribution in leukemic blasts. Quantitative analysis revealed significant correlation with proliferation markers, distinguishing normal from leukemic populations.
