The anti-mouse CD71 (TfR1) monoclonal antibody (clone R17217.1.3) was conjugated to HPMA copolymer-bound doxorubicin and injected intravenously at 5 mg doxorubicin equiv./kg in mice bearing 38C13 B-cell lymphoma. Tumor size was measured every 3 days using calipers, showing strong retardation of tumor growth. Survival was prolonged, with complete cures observed in three out of nine mice in the treated groups.

Anti-CD71 antibody-drug conjugate for the treatment of non-Hodgkin lymphoma

The anti-mouse CD71 (TfR1) antibody (clone R17217.1.3) was conjugated to monomethyl auristatin F and administered intravenously at 3 mg/kg to mice with Raji-luc tumors. Tumor burden was monitored weekly by bioluminescence imaging, showing complete tumor regression in all treated mice. Toxicity was assessed by body weight changes over 28 days, with no significant weight loss observed at therapeutic doses.

CD71-targeted therapy with anti-mouse CD71 antibody in a model of acute myeloid leukemia

The anti-mouse CD71 (TfR1) antibody (clone R17217.1.3) was injected intraperitoneally at 10 mg/kg twice weekly for 4 weeks in NSG mice engrafted with AML cells, depleting leukemic blasts. Bone marrow was analyzed by flow cytometry, showing an 80% reduction in human CD45+CD33+ cells. Overall survival was extended by 35 days compared to controls, demonstrating the antibody’s efficacy in AML treatment.

Iron oxide nanoparticles targeted to CD71 for magnetic resonance imaging of tumors in vivo

Anti-mouse CD71 (TfR1) antibody (clone R17217.1.3)-conjugated iron oxide nanoparticles were injected intravenously into tumor-bearing mice for MRI imaging. T1-weighted images acquired at 3T showed contrast enhancement in CD71-expressing tumors 24 hours post-injection. Biodistribution assessed by ICP-MS revealed 5-fold higher tumor accumulation compared to non-targeted nanoparticles, highlighting the antibody’s targeting efficiency.

Depletion of CD71+ erythroid cells with anti-mouse CD71 antibody in a mouse model of hemolytic anemia

The anti-mouse CD71 (TfR1) antibody (clone R17217.1.3) was administered intraperitoneally at 200 ?g per mouse on days 1 and 3 in phenylhydrazine-treated mice, depleting circulating reticulocytes. Reticulocyte counts were determined by flow cytometry using thiazole orange, and hematocrit levels were monitored daily for 7 days. Depletion reduced hemolysis-associated bilirubin levels by 50%, demonstrating the antibody’s efficacy in managing hemolytic anemia.
