The study utilized fluorochrome-conjugated monoclonal antibodies, including anti-human CD20 (clone B9E9), to stain human cells and analyze them via flow cytometry using Gallios or BD FACSymphony cytometers. Eight weeks post-injection in humanized mice, PBMCs were stained with anti-human CD45 to confirm humanization. Other antibodies included anti-CD3 (UCHT1), CD4 (SK3), CD8 (RPA-T8), and multiple others to assess immune cell profiles, demonstrating enhanced antitumor immunity through T-cell redirection.

For immunophenotyping, 50 µL of fresh heparinized peripheral blood (PB) or bone marrow (BM) was stained with antibodies including anti-CD19 (anti-human CD20, clone B9E9) for PB and BM analysis. Samples were acquired on Navios or Navios/EX cytometers, with cell populations identified based on forward scatter and CD45 expression. The study used a panel of antibodies, including CD3, CD7, CD5, CD34, and others, to gate lymphocytes, monocytes, granulocytes, and immature cells for risk stratification in AML and MDS.

CD20 expression in de novo diffuse large B-cell lymphoma was confirmed using the mouse anti-CD20 antibody B9E9 (anti-human CD20, IgG2a, recognizing the B1 epitope) via flow cytometry with a BD FACSAria III. The mean fluorescence intensity (MFI) of CD20 IHC(+)/FCM(−) cells was significantly lower than IHC(+)/FCM(+) cells when stained with B9E9 or B1 antibodies. CD19 positivity confirmed B-cell lymphoma populations, and CD20 IHC was performed with the L26 antibody.

The B9E9 scFvSA fusion protein, containing the single-chain variable region of the murine IgG2a anti-CD20 antibody B9E9 fused to streptavidin, was used to deliver HLA-A2/class I peptide complexes to HLA class I-negative Daudi cells. PBMCs were incubated with B9E9 scFvSA (10 μg/ml) for 1 hour, and flow cytometry with antibodies like anti-MHC class I (W6/32), anti-CD19, anti-CD80, and anti-CD86 confirmed binding and CTL induction.

A tetravalent single-chain antibody-streptavidin fusion protein for pretargeted lymphoma therapy

The B9E9 scFvSA fusion protein, derived from the CD20-specific murine IgG2a monoclonal antibody B9E9, was genetically engineered for pretargeted radioimmunotherapy. The V H and V L DNA fragments were amplified from B9E9 hybridoma cells, and the resulting scFvSA fusion protein showed nanomolar affinity to CD20-positive Ramos cells, comparable to the original B9E9 monoclonal antibody, as confirmed by in vitro and in vivo characterization.
