IgG-Based Bispecific Anti-CD95 Antibodies for the Treatment of B Cell-Derived Malignancies and Autoimmune Diseases.

The study utilized hybridoma-derived antibodies 2H7, 4G7, and Apo-1 for quantification. The bispecific antibodies (bsAbs) combined the CD95 agonist Apo-1 with either the anti-CD20 antibody clone 2H7 or the anti-CD19 clone 4G7 in F(ab)2 and single-chain formats. The CD95xCD20 bsAb demonstrated remarkable potency with IC50 values as low as 25 pM, despite the relatively low affinity of the anti-CD20 mAb clone 2H7 as a single chain. Expression of CD20, CD19, and CD95 on various cell lines was quantified using flow cytometry with QIFIKIT calibration beads, and binding of bsAbs to Daudi and Jurkat cells was assessed via flow cytometry.

In this study, female VH125.hCD20/NOD mice (6–8 weeks old) were treated with either anti-hCD20 antibody (clone 2H7) or control IgG2b antibody (clone MPC-11). Over a 12-week observation period, 48% of control-treated mice developed diabetes compared to 15% of 2H7-treated mice. The study involved injecting groups of 6- to 8-week-old VH125.hCD20/NOD mice with 2H7 anti-CD20 or IgG isotype control, followed by analysis of spleen and pancreatic lymph node (PLN) tissue for anti-insulin B cells post-treatment.

Targeted delivery of a PD-1-blocking scFv by CAR-T cells enhances anti-tumor efficacy in vivo.

The study employed flow cytometry to quantify PD-1 expression on CAR-T cells, with representative dot plots showing decreased PD-1 detection on 1928z-E27 and 4H1128z-E27 T cells compared to second-generation CAR-T cells. Western blot analysis on flow-sorted CAR+ and CAR− T cell populations was conducted using anti-HA mAb. A unique peptide (FGSNLESGIPAR, m/z = 624.3226) from the EH12/2H7 antibody was identified via Skyline analysis, confirming its absence in mouse or human proteomes, supporting its use in characterizing the 2H7 anti-human CD20 antibody.

Expression of lectin-like transcript-1 in human tissues.

The study evaluated a novel anti-LLT1 antibody (clone 2H7) for its staining capabilities in immunohistochemistry and flow cytometry, demonstrating effective recognition of lectin-like transcript-1 (LLT1) on germinal centre B cells. The 2H7 mAb, also an anti-human CD20 antibody, was used at a 1/500 dilution to stain human tissues (testes, brain, and placenta), proving its utility in studying LLT1 distribution through immunohistochemical staining.

Development and Characterization of a New Antipeptide Monoclonal Antibody Directed to Human CD20 Antigen.

This study developed a new monoclonal antibody, 3B4C10, targeting chimeric human CD20 extra loops (hCD20 exl), which showed strong reactivity with both peptide and native antigens on Raji B-cell lines. The antibody’s balanced K(on) and K(off) suggest potential applications in affinity columns or beads for isolating CD20-positive cells and cancer stem cells. Another antibody, C12H mAb, demonstrated high affinity and reactivity with native antigens on Raji and Daudi cells in flow cytometry, highlighting its potential for immunotherapy targeting the B-cell CD20 antigen.
