Description

Background

The anti-HEL VHH (hen egg-white lysozyme; nanobody, single-domain antibody, or the variable domain of heavy chain antibody) in nature is suitable for use as an isotype-matched control antibody to estimate the degree of non-specific binding by an antigen-specific antibody. As an ideal recombinant VHH isotype control antibody,  the recombinant anti-HEL VHH isotype control antibody has low or no specific binding to any human and mouse sample. The recombinant anti-HEL VHH isotype control antibodies are useful for in vitro and in vivo studies from ELISA, cell-based assay to animal model study.

Development of novel humanized VHH synthetic libraries based on physicochemical analyses.

This study focuses on heavy-chain antibodies (HCAbs), which lack the light chain and the first heavy chain constant region. The variable antigen-binding domain (VHH) of HCAbs, with a size of approximately 15 kDa, remains functional as a single domain. Novel synthetic humanized VHH libraries were designed and constructed using the complementarity determining region-grafting method and comprehensive sequence analyses of VHHs from the protein data bank. Due to their high affinity and specificity, VHHs are an attractive alternative to conventional fragment antibodies for various antibody-based applications.

The development of single-domain VHH nanobodies that target the Candida albicans cell surface.

This study measured the mean fluorescence intensity (MFI) for C. albicans als4? mutant and SC5314 strain, in various conditions, immunolabeled with VHH19, isotype control (#34), no VHH control, and detection antibody-only control. VHH libraries were panned against echinocandin-resistant C. albicans isolates and counter-selected against susceptible isolates to isolate binding domains specific for drug-resistant cells. Thirty diverse VHH nanobodies were selected, and their binding characteristics were assessed via dose-response ELISA, showing specificity for C. albicans across various isolates and other Candida species.

This study assembled paired consensus R1 and R2 reads into a single sequence spanning the VHH and part of the hinge region using the flash pairwise aligner. IGHV, IGHD, and IGHJ germlines were assigned using IgBlast with the IMGT-published alpaca reference germline set. Constant domain isotypes were identified using llama hinge-derived sequences with 100% sequence pattern matching. VHHs, or nanobodies, are single antigen-binding domains from camelid heavy-chain antibodies, widely used as diagnostic, research, and therapeutic tools.

First-in-class trispecific VHH-Fc based antibody with potent prophylactic and therapeutic efficacy against SARS-CoV-2 and variants.

This study evaluated the binding of ABS-VIR-001, a trispecific VHH-Fc antibody, to SARS-CoV-2 S RBD using high-binding 96-well plates coated with 1 ?g/ml of S RBD. ABS-VIR-001 and a VHH-Fc cocktail were serially diluted and tested in ELISA assay buffer. Binding and inhibition were assessed using a probe loaded with ACE-2-b and S protein (wild-type or mutants) in the presence or absence of ABS-VIR-001, with association curves generated by GatorPrime software to demonstrate qualitative binding and inhibition signals.
