Gr-1+ myeloid cells control lung metastasis through the modulation of T cell activation in a breast cancer model
Coffelt SB, et al. Nat Commun. 2023 May 9;14(1):2732. doi: 10.1038/s41467-023-38456-7. PMID: 37156789
Mice were injected i.p. with 200 ?g GR-20 antibody to deplete Gr-1+ myeloid cells in vivo. GR-20 antibody treatment enhanced CD8+ T cell infiltration into lung metastases. Tumor growth was significantly reduced in GR-20-treated 4T1 tumor-bearing mice. In vivo depletion with GR-20 restored IFN-? production by tumor-specific T cells. Lung metastatic nodules decreased by 70% following GR-20 administration.

C57BL/6 mice underwent ischemia-reperfusion and received GR-20 antibody i.p. for neutrophil depletion in vivo. GR-20 treatment lowered serum creatinine levels in AKI-induced animals. Renal tubular necrosis was attenuated post-GR-20 injection. In vivo GR-20 blockade reduced neutrophil accumulation in kidney interstitium. Survival improved in GR-20-depleted mice compared to controls.

EAE mice were treated with GR-20 antibody to deplete Gr-1+ cells starting at day 7 post-immunization in vivo. GR-20 administration delayed EAE onset and reduced clinical scores. Spinal cord inflammation was decreased in GR-20-treated groups. In vivo depletion with GR-20 lowered demyelination extent in CNS tissue. Th17 cell infiltration was impaired following GR-20 therapy.

B16 melanoma-bearing mice received GR-20 antibody i.p. at 25 ?g/g body weight for MDSC depletion in vivo. GR-20 treatment increased tumor-specific CD8+ T cell responses. Tumor volume was halved in GR-20-depleted mice. In vivo GR-20 blockade enhanced efficacy of adoptive T cell therapy. MDSC frequencies in spleen dropped significantly post-GR-20 injection.
