CSF1R inhibition reprograms tumor-infiltrating macrophages and improves response to T cell checkpoint immunotherapy in a mouse model of breast cancer

Cells were stained with anti-mouse CD115 (clone AFS98, BioLegend) conjugated to PE for flow cytometric analysis of CSF1R expression on tumor-associated macrophages. Tumor-infiltrating leukocytes were isolated from digested breast tumors and stained for flow cytometry. Flow cytometry was performed on a BD FACSCanto II with compensation using single-stained controls. CSF1R inhibition reduced CD115+ F4/80+ TAM frequency by 40%. Gating strategy focused on live CD45+ CD11b+ Ly6G- cells.

Flow cytometric analysis of CSF1R (CD115) on mouse monocytes using clone AFS98

Monocytes were labeled with anti-mouse CD115 (AFS98 clone, eBioscience) for multicolor flow cytometry. Samples were acquired on a LSRFortessa flow cytometer and analyzed with FlowJo software. Data analysis included compensation and viability gating. CD115 expression was highest on Ly6C+ monocytes. Stimulation with M-CSF upregulated surface CD115 levels.

Role of CD115 in macrophage activation assessed by flow cytometry with AFS98 antibody

Bone marrow-derived macrophages were stained with anti-CD115 (clone AFS98) to monitor receptor expression by flow cytometry. Cells were fixed and permeabilized for intracellular cytokine staining. FACS analysis showed upregulation during LPS stimulation. CD115+ cells correlated with TNF-α production. Controls included isotype-matched antibodies.

CSF1R expression on dendritic cells by flow cytometry using clone AFS98 in a mouse model

Myeloid cells from spleen were analyzed using anti-mouse CD115 (AFS98) in flow cytometry. Viable cells were selected using propidium iodide exclusion. Expression changes were quantified under inflammatory conditions. CD115 MFI increased significantly in LPS-treated samples. Gating strategy focused on CD11c+ and CD11b+ subsets.

Flow cytometry-based detection of CD115 on myeloid cells with anti-CSF1R clone AFS98

Dendritic cells were stained with anti-CSF1R (clone AFS98) for flow cytometric quantification. Phagocytosis was assessed by uptake of fluorescent beads. BD LSR II was used for data acquisition. Mature DCs showed higher CD115 expression. Blocking with AFS98 reduced phagocytic activity.
