Description

Background

The anti-DNP antibodies specifically bind to DNP (2,4,6-trinitrophenyl) and derivatives. The DNP antibodies have been used in various applications, such as ELISA, Immunohistochemistry, Immunocytochemistry, and Western Blot. Because Dinitrophenyl (DNP) is a hapten and not found endogenously in animal and plant tissues, the anti-DNP monoclonal antibody in nature is suitable for use as an isotype-matched control antibody to estimate the degree of non-specific binding by an antigen-specific antibody. The recombinant mouse IgG1 kappa isotype control (clone number: DNP.m1) and mutants (natural and engineered mIgG1 isotype controls) are useful for in vitro and in vivo studies from ELISA, Western blot (WB), to cell-based assay to animal model study. The isotypes of mouse IgG antibody reagents used in scientific research and clinical diagnostics are mainly mouse IgG1 kappa. Mouse IgG1 antibodies and engineered mutants are also used in therapeutic drug discovery. Mouse IgG1 antibodies are more like human IgG4 antibodies than human IgG1 antibodies, which behave more like mouse IgG2a or IgG2c antibodies. Many antibody drugs especially those targetting immune checkpoint proteins use the human IgG4 constant regions.

As an ideal recombinant mouse IgG1 isotype control,  the recombinant anti-DNP mIgG1 kappa isotype control has low or no specific binding to any mouse sample, and has completely mouse variable regions and the mouse IgG1 kappa constant regions. The anti-DNP mouse IgG1 kappa isotype control and mutants can be used in ELISA, WB, IHC, the cell-based assay or animal model as the antibody negative control.

Antibody Isotype Switching as a Mechanism to Counter HIV Neutralization Escape.

This study demonstrates that IgG3 and IgA1 isotypes exhibit superior neutralization of longitudinal autologous HIV viruses and epitope mutants compared to IgG1. Additionally, it highlights that detrimental class-switch recombination (CSR) events leading to reduced neutralization can be reversed through further CSR, termed "switch redemption." CSR thus serves as an additional immunological mechanism to counter viral escape from HIV-specific antibody responses. The study also notes that somatic hypermutation (SHM) in immunoglobulin variable regions enables maturing antibodies to neutralize emerging viral escape variants, while the Ig-constant region, determining isotype, influences epitope recognition.

A universal dual mechanism immunotherapy for the treatment of influenza virus infections.

This study evaluates the ability of zan-DNP to recruit anti-DNP antibodies to influenza A(H1N1)-infected cells, using biotinylated anti-DNP antibodies for visualization on MDCK cells. Confocal micrographs confirmed that anti-DNP antibodies bind only to infected cells treated with zan-DNP. In vivo, mice challenged with 100x MLD50 of A/California/07/2009 (H1N1)pdm09 virus and treated with 0.5 ?mol/kg zan-DNP or zanamivir plus anti-DNP antibodies (5 mg/kg, i.v.) showed that 24.4% of infected lung cells were opsonized with antibody in zan-DNP-treated mice. The study also tested co-injection of exogenous anti-DNP antibodies to compensate for low endogenous anti-DNP antibody titers, demonstrating an effective anti-influenza immune response.

Hapten-mediated recruitment of polyclonal antibodies to tumors engenders antitumor immunity.

This study investigates the use of VEGF-DNP hapten conjugates to recruit polyclonal antibodies to tumors, demonstrating that polyclonal antibodies are more effective than monoclonal anti-DNP antibodies in eliciting tumor immunity. Experiments showed that VEGF-DNP, but not control aptamer-DNP conjugates, inhibited tumor growth in DNP-immune (DNP-KLH) mice but not in mock (KLH)-vaccinated mice. The VEGF-DNP conjugate incubated with monoclonal anti-DNP or isotype control antibody (D8406, SIGMA-Aldrich) at a 2:1 ratio was largely ineffective. Statistical analysis showed significant tumor inhibition with VEGF-DNP in DNP-KLH mice (p = 0.0066, Student’s t-test, n = 1).

Antibodies with idiotypic and anti?idiotypic reactivity (epibodies) in conventional immune responses to dinitrophenylated carriers.

This study found that only 7% of hybrid supernatants from spleens undergoing a primary response to DNP-Ba exhibited both Ab1 and Ab2 reactivity. After cloning, monoclonal antibodies (mAbs) were characterized for isotype and reactivity to DNP-BSA, rabbit anti-DNP antibodies, and unrelated antigens. Approximately half of the mAbs from the primary response to DNP-Ficoll were IgM and polyreactive. All mAbs with dual reactivity from DNP-Ba responses were also polyreactive and belonged to the IgM class, fitting the definition of epibodies with specificity for both the hapten and anti-hapten antibody.

125 I-labeled anti-2, 4-dinitrophenyl (DNP)-antibodies: A universal reagent for solid phase radioimmunoassays.

This study describes the development of solid phase radioimmunoassays (RIA) using 125I-labeled anti-DNP antibodies to quantify antigen-bound antibodies. The method employs dinitrophenylated antibodies, which are detected by the attachment of 125I-labeled anti-DNP, offering advantages such as using a single radiolabeled antibody for all tests, amplified tracer binding, and eliminating the need for immunochemical purification of each antibody. This approach allows haptenated immunoglobulins to serve as a general tracer in solid-phase RIA.
