Description
p>Normal mouse serum and purified mouse IgGs from normal mouse serum are commonly used as non-specific passive blocking solution for antibody-based immuno-assays. Recombinant mouse IgG antibody mixes, as a direct alternative to normal mouse serum and even mouse IgGs purified from mouse serum or a non-specific passive blocking solution, can be used to eliminate interfering antibodies from immuno-assay systems using capture and detection antibodies. There is higher percentage of mouse IgG1 or IgG2b in serum from different strains of mice. So, recombinant mouse IgG antibody mixes with different percentages of mouse IgG1, IgG2a, IgG2b, IgG2c, and IgG3 were made to meet customers' requirements of different applications. A customized recombinant mouse IgG antibody mix with different percentages of mouse IgG1, IgG2a, IgG2b, IgG2c, and IgG3 can be provided.

Features of recombinant mouse IgG antibody mixes compared with normal mouse serum and serum-derived mouse IgGs:

1) Animal free production. CHO cells rather than mice are used in antibody production.

2) Lot-to-lot consistency. CHO cells which are used for antibody drug manufacturing are used to manufacture the mouse IgG antibodies in the antibody mixes. All production is under control and predictable.

3) As a targeted or passive antibody blocker to interfering antibodies like HAMA (Human Anti-Mouse Antibodies) and no binding to human tissues. In contrast, some serum-derived mouse IgGs may bind to unknown targets in some human tissues.

4) Adjustable percentages of the antibodies in the antibody mixes. A customized recombinant mouse IgG antibody mix with different percentages of mouse IgG1, IgG2a, IgG2b, IgG2c, and IgG3 can be easily made according to customers' applications.

Question: Normal mouse serum is often used for blocking or saturating immunoassays, especially those involving tissue samples such as immunohistochemistry (IHC). How much should recombinant antibody mixes be added to replace normal mouse serum?
Answer: Our recombinant mouse IgG antibody mixes can be used as normal mouse serum alternative for blocking reagent, and more specifically, the purified normal IgGs alternative. It was said that there is 80 to 90 mg/ml protein in a normal mouse serum sample, and 7 to 10 mg/ml IgG in a typical mouse serum and plasma sample. It means that there is 7.8% to 12.5% IgGs in proteins in normal mouse serum. One may calculate the amount of mouse IgGs in a mouse serum sample. One should also consider the percentage of various mouse IgG isotypes in the mouse serum sample vs recombinant mouse IgG antibody mixes. Conditions of optimal mouse IgG antibody mix performance should be determined experimentally by customers.

The most popular human Fc blocking reagents include:
Anti-human CD16 (clone 3G8), and CD32 (Clone IV.3): for flow cytometry, immunohistochemistry (IHC), and immunoprecipitation (IP). It is highly specific for FcγRII (CD32) and FcγRIII (CD16), and reduces background staining; it may not block all Fc receptors. Clones 10.1, and 22 or H22 are the most widely used anti-human FcγRI (CD64) clone for Fc blocking in flow cytometry and functional assays. It prevents non-specific Fc receptor binding on monocytes, macrophages, and dendritic cells, improving antibody specificity.
Purified human IgGs or mixes of human isotype controls: for general Fc blocking in IHC and immunofluorescence (IF). It is cheap and easy to use but it is less specific, and may even introduce unwanted immunoglobulins. Syd Labs supply various human IgG isotype controls.
Normal mouse serum, purified mouse IgG or isotype controls: for flow cytometry. It can serve as a control to block Fc receptors but it is non-specific for Fc receptors. It is cheap and easy to use but it is less specific, and may even introduce unwanted immunoglobulins. Bovine Serum Albumin (BSA) or FBS: for general blocking in Western Blot (WB) and IHC. They are readily available to reduce non-specific binding but less effective at Fc receptor blocking.
Commercial Fc blocking solutions: for flow cytometry, IHC, and functional assays. They are normally pre-optimized and highly effective but more expensive than DIY solutions. 

Best choice of human Fc blocking reagents based on your application are:
For flow cytometry: anti-human CD16 (Clone 3G8), CD32 (Clone IV.3), and CD64 (Clone 10.1, and 22 or H22).
For Immunohistochemistry (IHC) and Immunofluorescence (IF): Normal mouse serum, purified mouse or human IgGs or isotype controls or commercial Fc blocking kits. A novel mouse strain optimized for chronic human antibody administration

This model combines both a human IgG1 heavy chain knock-in and a full recapitulation of human Fc receptor (FcγR) expression, providing a unique platform for in vivo testing of human monoclonal antibodies with relevant receptors beyond the short term. Compared to controls, hIgG1 knock-in mice mount minimal anti-human IgG responses, allowing for the persistence of therapeutically active circulating human IgG even in the late stages of treatment in chronic models of immune thrombocytopenic purpura and metastatic melanoma. Therapeutic human IgG antibodies are routinely tested in mouse models of oncologic, infectious, and autoimmune diseases. IgG modulation in male mice with reproductive failure after prenatal inflammation

The immunomodulatory effect of mouse IgG on reproductive maturity in male offspring after LPS exposure in vivo and in vitro was studied. Prenatal IgG injection (20 µg/mouse) had a positive impact on the puberty of male mice whose mothers were exposed to LPS (100 µg/kg) on the 12th day of pregnancy. The numbers of Sertoli cells were increased, whereas the body weight and the number of symplastic spermatids were decreased in offspring as compared to LPS-treated animals. Besides, IgG had a positive effect on altered hormone levels: reduced estradiol level on the 5th and 14th postnatal days and increased testosterone level on the 30th postnatal day in blood. This led to an increased number of mounting attempts in sexually mature males.

A Monosaccharide Residue Is Sufficient to Maintain Mouse and Human IgG Subclass Activity and Directs IgG Effector Functions to Cellular Fc Receptors

However, this study reveals that a mono- or disaccharide structure consisting of one N-acetylglucosamine with or without a branching fucose residue is sufficient to retain the activity of the most active human and mouse IgG subclasses in vivo. Immunoglobulin G (IgG) glycosylation modulates antibody activity and represents a major source of heterogeneity within antibody preparations. Depending on their glycosylation pattern, individual IgG glycovariants present in recombinant antibody preparations may trigger effects ranging from enhanced pro-inflammatory to anti-inflammatory activity. FcgammaRIV: a novel FcR with distinct IgG subclass specificity

Mouse IgG subclasses display a hierarchy of in vivo activities, with IgG2a and IgG2b showing the greatest protective and pathogenic properties. These enhanced activities result, in part, from their ability to bind to a novel, gamma chain-dependent, activating IgG Fc receptor, FcgammaRIV. Thus, the FcgammaR system has evolved distinct activation receptors displaying selectivity for IgG subclasses, with IgG1 antibodies exclusively dependent on FcgammaRIII, whereas IgG2a and IgG2b show preferential dependence on FcgammaRIV activation. These distinct binding affinities for the IgG subclasses to FcgammaRs account for their differential protective and pathogenic activities in vivo. Wild-type and FcgammaRIV-deficient mice were challenged intravenously with IgG-opsonized red blood cells.
