BP005345: Human PD-L1/B7-H1 Protein with the C-His Tag

Source: The recombinant human PD-L1/B7-H1 protein, the PD-L1 ECD (amino acids Phe19-Arg238) fused with a His-tag at the C-terminus, was produced in HEK 293 cells.
Molecular Weight: The recombinant human PD-L1/B7-H1 protein with the C-His tag has a predicted MW of 26 kDa. Due to glycosylation, the protein migrates to 35-42 kDa based on Tris-Bis PAGE result.
Activity: The EC50 of the anti-human PD-L1 antibody with the hFc tag to the immobilized human PD-L1/B7-H1 protein with the His tag at 0.5 ug/ml (100 ul/Well) was 6.4 ng/ml determined by ELISA.
Purity: >95% by SEC-HPLC and SDS-PAGE under reducing conditions.
Endotoxin Level: Less than 1 EU per ug of protein as determined by LAL method.
Formulation: Lyophilized from 0.22 um filtered solution in PBS (pH 7.4). Normally 8% trehalose is added as protectant before lyophilization.
Reconstitution: It is recommended to reconstitute the lyophilized recombinant human PD-L1/B7-H1 protein in sterile H2O to a protein concentration not less than 100 ug/ml. Centrifuge the tube before opening.

Molecular Weight: The recombinant human PD-L1/B7-H1 protein with the C-His tag has a predicted MW of 24.9 kDa. Due to glycosylation, the protein migrates to 35-45 kDa based on Tris-Bis PAGE result.
Activity: The EC50 of the anti-human PD-L1 antibody with the hFc tag to the immobilized human PD-L1/B7-H1 protein with the His tag at 0.5 ug/ml (100 ul/Well) was 4.3 ng/ml determined by ELISA.

Activity: The EC50 of the recombinant human PD-L1 protein with the hFc tag to the immobilized human PD-1/PDCD1  with the C-mFc tag at 1 ug/ml (100 ul/Well) was 0.17 ug/ml determined by ELISA. The recombinant human PD-L1 with the C-hFc Tag captured on CM5 Chip via Protein A can bind to the recombinant human PD-1 with the C-His Tag with an affinity constant of 2.20 uM as determined in SPR assay (Biacore T200).
