Background
The 1F4 antibody binds to the TIGIT protein (WUCAM, Vstm3 or VSIG9), a novel immune checkpoint receptor with inhibitory function. TIGIT is expressed on T cells and natural killer (NK) cells, and several human cancers, including melanoma, NSCLC, and colorectal cancer. Similar to PD-1, the TIGIT receptor limits antitumor immune response in cancer.

NS1-specific B cells were sorted using 1F4 (anti-mouse TIGIT antibody) staining. Antibody responses were assessed by ELISPOT assays. Flow cytometry was performed on a BD LSRFortessa.

A humanized monoclonal antibody specific for invariant Natural Killer T (iNKT) cells for in vivo depletion.

The chimeric 1F4 antibody (anti-mouse TIGIT) was generated by combining the variable regions of the mouse anti-human iNKT TCR antibody (clone 6B11) with human IgG1 constant regions. Mice were injected intraperitoneally with 1F4 (anti-mouse TIGIT mAb) to deplete iNKT cells. Flow cytometry confirmed iNKT cell depletion using 6B11 staining. Depletion efficacy was assessed in spleen and liver. In vivo studies showed reduced iNKT cell-mediated cytokine production.

The 1F4 antibody (anti-mouse TIGIT) was used to stain EGFR in drug-tolerant cancer cells for flow cytometry. PC9 cells were treated with erlotinib in vivo in xenograft models. Tumor volumes were measured weekly by caliper. Flow cytometry analysis showed persistent EGFR expression in drug-tolerant cells. Drug-tolerant persisters were isolated for further genomic analysis.
