Background
The 10A7 antibody binds to the TIGIT protein (WUCAM, Vstm3 or VSIG9), a novel immune checkpoint receptor with inhibitory function. TIGIT is expressed on T cells and natural killer (NK) cells, and several human cancers, including melanoma, NSCLC, and colorectal cancer. Similar to PD-1, the TIGIT receptor limits antitumor immune response in cancer.

To determine whether the NRP2+ neuroblastic progenitor state could be targeted for immunotherapy, we combined anti-TIGIT antibody clone 10A7 with anti-PD-L1 in syngeneic neuroblastoma mouse models. Single-cell suspensions were stained with fluorochrome-conjugated antibodies for flow cytometry. Tumor growth was monitored by caliper measurements twice weekly. The combination therapy significantly reduced tumor burden compared to controls. Flow cytometry data were analyzed using FlowJo software.

The immune checkpoint TIGIT is upregulated on T cells during infection and downregulates T cell function to limit antibacterial immunity.

Anti-TIGIT antibody (clone 10A7) was administered intraperitoneally to block TIGIT during Salmonella infection. Flow cytometry confirmed upregulation of TIGIT on CD8+ T cells using clone 10A7 (anti-mouse TIGIT antibody). Infected mice were monitored for bacterial burden in spleen and liver. The blockade enhanced antibacterial T cell responses. Data were acquired on a BD LSRFortessa flow cytometer.

Enhanced antitumor efficacy of a novel oncolytic vaccinia virus encoding a fully monoclonal antibody against TIGIT.

The oncolytic vaccinia virus was engineered to express a single-chain variable fragment (scFv) derived from the anti-TIGIT antibody clone 10A7. In vivo studies were conducted in BALB/c mice bearing CT26 tumors. Tumor volumes were measured every 3 days post-treatment. The engineered virus significantly prolonged survival compared to control virus. Flow cytometry confirmed increased CD8+ T cell infiltration in treated tumors.

An engineered oncolytic vaccinia virus encoding a fully monoclonal antibody against TIGIT induces effective antitumor responses through increased M1 macrophage recruitment.

The virus encodes the heavy and light chains of anti-TIGIT antibody (clone 10A7) connected by a self-cleaving 2A peptide. Tumor-bearing mice were treated intratumorally with the engineered virus. Flow cytometry was used to analyze macrophage polarization using anti-F4/80 and anti-CD86. The treatment increased M1 macrophage recruitment in the tumor microenvironment. Survival was significantly improved in the treatment group.

The anti-TIGIT antibody 10A7 was used to stain human T cells for flow cytometry to assess TIGIT expression. Crystal structures were determined for TIGIT bound to PVR. In vivo blocking experiments with 10A7 (anti-mouse TIGIT mAb) enhanced T cell responses in mouse models. Flow cytometry data showed 10A7 (anti-mouse TIGIT antibody) binding to TIGIT-transfected cells. The cis-trans regulatory mechanism was validated in cell adhesion assays.
