Background
The 1F4 antibody binds to the TIGIT protein (WUCAM, Vstm3 or VSIG9), a novel immune checkpoint receptor with inhibitory function. TIGIT is expressed on T cells and natural killer (NK) cells, and several human cancers, including melanoma, NSCLC, and colorectal cancer. Similar to PD-1, the TIGIT receptor limits antitumor immune response in cancer.

The amyloidoma is readily visible (arrow) and anatomically pinpointed when a 3-D surface rendering of the 125I-11-1F4 mAb distribution is displayed with the CT image. We have developed microimaging technologies, implemented novel computational methods, and established protocols to generate high-resolution images of amyloid deposits in mice. Typical biodistribution data following 125I-SAP injection into mice with systemic AA amyloidosis. H-2/IL-6 mice (with amyloid) and wild type, healthy mice (WT) received an iv injection of 125I-SAP, were sacrificed 24 hr post-injection, and the organs harvested for analysis. Immunotherapy in systemic primary (AL) amyloidosis using amyloid-reactive monoclonal antibodies.

The chimeric 11-1F4 mAb was produced in CHOdhfr-stable mammalian cell lines that had been transfected with a supervector DNA encoding the genes for both the light and heavy chains. The c11-1F4 mAb was purified from culture supernatants by protein A affinity chromatography. Groups of 5 mice were injected subcutaneously (sc) with 100 mg of human AL amyloid extract on days 0, 3, 7, and 10, to induce the formation of an amyloidoma. On day 14, the mice received a single iv injection of 100 μg of the c11-1F4 mAb, or an isotype-matched control antibody (c2A4). The amyloidoma is readily visible (arrow) and anatomically pinpointed when a 3-D surface rendering of the 125I-11-1F4 mAb distribution is displayed with the CT image.
