Generation and characterization of monoclonal antibodies specific for mouse V?4 TCR

The UC3-10A6 antibody was used for in vivo depletion of V?4+ ?? T cells in an experimental psoriasis model, administered intraperitoneally at 200 ?g every three days. Skin inflammation was scored based on erythema, scaling, and thickness, with depletion efficiency confirmed by flow cytometry in skin lesions. UC3-10A6 treatment reduced imiquimod-induced psoriasis severity by 50%.

Ear thickness was measured daily with a micrometer, and histological analysis showed reduced epidermal hyperplasia. Cytokine levels, including IL-17 and IL-22, were quantified by ELISA, confirming UC3-10A6’s impact on psoriasis severity.

Role of V?4+ ?? T cells in Klebsiella pneumoniae infection
