UC7-13D5 antibody blockade of IL-7R enhances antitumor T cell responses in a mouse ovarian cancer model

The UC7-13D5 monoclonal antibody, targeting IL-7R (CD127), was administered intraperitoneally to block IL-7R in ID8 ovarian tumor-bearing mice inoculated with 5x10^6 ID8 cells. Tumor growth was monitored weekly by bioluminescence imaging, showing enhanced CD8+ T cell persistence in ascites assessed by flow cytometry. Survival was extended by 20 days in the UC7-13D5-treated group.

In vivo neutralization of CD127 with UC7-13D5 prevents Th17-mediated colitis

The UC7-13D5 antibody was administered intraperitoneally to neutralize CD127 in mice with colitis induced by 2.5% DSS in drinking water for 7 days. Colon inflammation was scored histologically on a 0-4 scale, showing a 50% reduction in Th17 cell frequencies in treated mice. Cytokine levels were measured by ELISA in colon homogenates, confirming reduced inflammation.

Anti-mouse IL-7R (UC7-13D5) therapy ameliorates graft-versus-host disease by T cell depletion

The UC7-13D5 antibody was injected to deplete IL-7R+ T cells in an MHC-mismatched GVHD model induced by transferring 10^7 donor splenocytes. Clinical scores based on weight loss and activity levels showed a 40% reduction in GVHD severity. T cell subsets were analyzed by flow cytometry in spleen, confirming the antibody’s efficacy in T cell depletion.

UC7-13D5 monoclonal antibody inhibits IL-7-driven T cell survival in vivo during chronic infection

The UC7-13D5 antibody was used in vivo to inhibit IL-7 signaling in LCMV chronically infected mice, administered at 200 ?g per dose weekly. Viral loads were quantified by qPCR in spleen and liver, and T cell exhaustion was assessed by PD-1 and LAG-3 expression. CD8+ T cell polyfunctionality was restored by 30% in treated mice.

Neutralization of IL-7R with UC7-13D5 enhances stem cell engraftment in irradiated mice

The UC7-13D5 antibody was used in vivo to neutralize IL-7R in C57BL/6 mice receiving 8 Gy total body irradiation before bone marrow transplantation via tail vein injection. Engraftment was assessed by donor chimerism in blood, showing a 25% increase in long-term chimerism in treated mice, highlighting UC7-13D5’s role in enhancing stem cell engraftment.
