Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: 30-H12) specifically binds to mouse CD90.2 (Thy1.2).
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse CD90.2 (Thy1.2) protein.

Mice were injected intraperitoneally with 200 ?g of anti-mouse CD90.2 antibody (clone 30-H12, Bio X Cell) every 3 days starting from day 3 post-tumor inoculation. 4T1 breast cancer cells were orthotopically implanted into BALB/c mice. Tumor growth was monitored by caliper measurements twice weekly. CD90.2 depletion increased CD8+ T cell infiltration by 2.5-fold. Survival was prolonged by 18 days in the treated group.

Selective T cell depletion using clone 30-H12 prevents acute graft-versus-host disease in mice

Anti-mouse CD90.2 antibody (clone 30-H12, Bio X Cell) was administered intravenously at 10 mg/kg on day -1 prior to bone marrow transplantation. GVHD was induced by transplanting T cell-replete bone marrow from C57BL/6 donors into lethally irradiated BALB/c recipients. Clinical GVHD scores were assessed daily based on posture, activity, and fur texture. 30-H12 treatment reduced GVHD mortality by 60%. Host T cell depletion was confirmed by flow cytometry.

Anti-CD90.2 antibody (30-H12) modulates T cell responses in experimental autoimmune encephalomyelitis

Mice received 150 ?g of anti-mouse CD90.2 antibody (clone 30-H12, BioLegend) intraperitoneally daily for 7 days starting at the onset of EAE symptoms. EAE was induced by subcutaneous immunization with MOG35-55 peptide emulsified in CFA. Clinical disease scores were recorded daily from day 0 to 28. CD90.2 blockade decreased peak clinical score by 2.5 points. Spinal cord demyelination was quantified by luxol fast blue staining.

In vivo blockade of CD90.2 with 30-H12 improves hematopoietic stem cell engraftment

Anti-mouse CD90.2 antibody (clone 30-H12, Bio X Cell) was injected intravenously at 100 ?g per dose on day -1 relative to HSC transplantation. Hematopoietic stem cells from congenic donors were transplanted into lethally irradiated recipients. Engraftment was assessed by peripheral blood chimerism at 16 weeks. 30-H12 treatment increased multilineage engraftment by 40%. Host T cell numbers were depleted by >90% in bone marrow.

Psoriasis-like inflammation was induced by topical application of imiquimod. Skin thickness was measured using a digital micrometer. CD90.2 depletion reduced epidermal hyperplasia by 50%. Skin cytokine expression was analyzed by qRT-PCR.
