Source: The anti-mouse CD47 monoclonal antibody (clone: Miap301) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: Miap301) specifically binds to mouse CD47.
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse CD47 protein.
Form of Antibody: 0.2 uM filtered solution, pH 7.4, no stabilizers or preservatives.

B16F10 melanoma-bearing mice were treated with anti-mouse CD47 antibody (miap301) i.p. to block CD47 in vivo. Miap301 administration enhanced macrophage phagocytosis of tumor cells in vivo. In vivo miap301 blockade reduced tumor growth in syngeneic models. Combination with PD-L1 antibody amplified miap301 effects on survival. Tumor rechallenge showed memory responses post-miap301 therapy.

Anti-CD47 Antibody As a Targeted Therapeutic Agent for Human Lung Cancer and Cancer Stem Cells

NOD/SCID mice engrafted with lung cancer cells received anti-mouse CD47 MIAP301 antibody i.p. for in vivo therapy. MIAP301 treatment inhibited tumor growth in xenografts. In vivo MIAP301 enhanced macrophage-mediated clearance of CSCs. Survival was prolonged in MIAP301-treated tumor-bearing mice. No significant toxicity was observed post-MIAP301 administration.

B16F10 melanoma-bearing mice were injected with anti-mouse CD47 antibody (Miap301) to disrupt SIRPα signaling in vivo. Miap301 treatment potentiated antibody-mediated tumor destruction. In vivo Miap301 blockade reduced melanoma lung metastases. Macrophage infiltration increased in Miap301-treated tumors. Therapeutic efficacy was enhanced by Miap301 in vivo.

CD47 blockade triggers T cell-mediated destruction of immunogenic tumors

Tumor-bearing mice received anti-mouse CD47 antibody (Miap301 clone) i.p. to activate T cells in vivo. Miap301 triggered T cell infiltration into tumors. In vivo Miap301 blockade destroyed immunogenic tumors. Tumor rechallenge resistance developed post-Miap301. CD8+ T cells were essential for Miap301 efficacy in vivo.

Targeting CD47-SIRPα as an adjunct to tumor immunotherapy

MC38 colon carcinoma mice were treated with anti-mouse CD47 (Miap301) to synergize with immunotherapy in vivo. Miap301 enhanced PD-1 blockade antitumor effects. In vivo Miap301 increased dendritic cell cross-presentation. Tumor growth was halted in Miap301 combination therapy. Immune memory was induced by Miap301 in vivo.
