The anti-CD47 antibody B6H12 suppressed tumor cell growth, motility, and promoted macrophage-mediated phagocytosis in IOMM-Lee meningioma cells in vitro. In vivo, B6H12 and MIAP301 significantly inhibited tumor growth in xenograft models, an effect partially blocked by macrophage depletion. Wound healing and transwell invasion assays confirmed reduced migration and invasion in B6H12-treated cells.

A function-blocking CD47 antibody suppresses stem cell and EGF signaling in triple-negative breast cancer

The B6H12 antibody inhibited tumor growth in triple-negative breast cancer xenograft models by preventing SIRPα engagement. In MDA-MB-231 and bCSCs cells, 24-hour B6H12 treatment significantly reduced cell proliferation (p<0.05), as shown by BrdU staining and flow cytometry analysis of KLF4 expression, indicating suppression of stem cell and EGF signaling.

Anti-CD47 monoclonal antibody (B6H12) impairs the maturation and function of human dendritic cells

The B6H12 monoclonal antibody, when present during monocyte-derived dendritic cell (DC) propagation with GM-CSF, LPS, and IL-4, impaired DC maturation and function by inhibiting NF-kappaB DNA binding activity. Flow cytometry, mixed leukocyte reaction, and IL-12 p70 production data (p<0.01) confirmed B6H12’s negative effect on DC development.

Effect of soluble anti-CD47 monoclonal antibody on the differentiation and function of human dendritic cells

The soluble B6H12 anti-CD47 antibody negatively impacted the maturation and function of human dendritic cells cultured with GM-CSF, LPS, and IL-4. Flow cytometry, semi-quantitative RT-PCR, and ELISA showed reduced IL-12 expression and lower BrdU incorporation in B6H12-treated DCs (p<0.01) compared to untreated DCs.

A Novel Blockade CD47 Antibody With Therapeutic Potential for Cancer

The B6H12.2 anti-CD47 antibody was used in flow cytometry to assess CD47 expression in 3T3-WT, 3T3-CD47 (GFP), Jurkat-shCON (GFP), and Jurkat-shCD47 (GFP) cell lines. Immunofluorescence via confocal microscopy confirmed binding after overnight incubation, demonstrating B6H12.2’s potential for therapeutic applications in cancer models.
