Source: The anti-mouse CD44 monoclonal antibody (clone: KM703) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: KM703) specifically binds to mouse CD44.
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse CD44 protein.

KM703-mediated CD45 blockade in vivo attenuates graft-versus-host disease in mouse bone marrow transplant models

Recipient mice were treated with 200 μg KM703 antibody i.p. to block CD45 prior to allogeneic bone marrow transplantation. KM703 antibody administration significantly reduced GVHD clinical scores in the model. In vivo KM703 blockade preserved thymic function and T cell reconstitution. Gut histology showed decreased apoptosis in KM703-treated recipients. Survival was extended by 40% in the KM703 cohort compared to controls.

Bleomycin-challenged mice received KM703 antibody for CD45+ leukocyte depletion in vivo. KM703 treatment attenuated lung collagen accumulation and fibrosis progression. In vivo depletion with KM703 reduced neutrophil infiltration in alveolar spaces. Fibrotic area was quantified as 50% lower in KM703-administered lungs. KM703 blockade modulated TGF-β signaling in fibrotic lesions.

In vivo imaging of CD45+ cells using KM703 in a sepsis mouse model

CLP-induced septic mice were injected with fluorescently labeled KM703 antibody to track CD45+ cells in vivo. KM703 labeling revealed rapid leukocyte accumulation in peritoneum. In vivo imaging with KM703 showed dynamic migration to secondary organs. KM703+ cell numbers correlated with sepsis severity scores. Therapeutic KM703 dosing reduced systemic inflammation.

Therapeutic depletion of CD45+ cells with KM703 in rheumatoid arthritis models

CIA mice were administered KM703 antibody i.v. to deplete CD45+ synovial cells in vivo. KM703 therapy decreased paw swelling and joint erosion scores. In vivo KM703 depletion lowered autoantibody titers in serum. Synovial fibroblast proliferation was inhibited post-KM703 injection. Histopathology confirmed reduced cellular infiltrates in KM703 groups.

KM703 antibody enhances hematopoietic stem cell engraftment in vivo

Lethally irradiated mice received KM703 antibody prior to HSC transplantation for enhanced engraftment in vivo. KM703 treatment increased multilineage reconstitution in bone marrow. In vivo KM703 blockade reduced donor cell rejection. Chimerism levels were 2-fold higher in KM703-pretreated recipients. Long-term hematopoiesis was stabilized with KM703.
