Bone marrow-derived DCs were stained with anti-mouse CD370 (clone 10B4, BioLegend) conjugated to PE for flow cytometry analysis. Cells were gated on live CD11c+ MHC II+ populations. Flow cytometry was performed using a BD FACSCanto II instrument. Dectin-1 expression increased 2-fold upon beta-glucan stimulation. Cytokine production was measured by intracellular staining post-stimulation.

Flow cytometric analysis of Dectin-1 (CD370) on mouse macrophages using clone 10B4

Macrophages were labeled with anti-mouse CD370 (10B4 clone, eBioscience) for multicolor flow cytometry. Samples were acquired on a LSRFortessa flow cytometer. Data analysis was conducted using FlowJo software. CD370 expression was highest on F4/80+ CD11b+ cells. Stimulation with zymosan upregulated surface CD370 levels.

NK cells and DCs were stained with anti-CD370 (clone 10B4) to monitor receptor expression by flow cytometry. Cells were fixed and permeabilized for intracellular markers. FACS analysis showed dynamic upregulation during infection. Dectin-1 positive cells correlated with IL-12 production. Controls included isotype-matched antibodies.

Dectin-1 expression on myeloid cells by flow cytometry using clone 10B4 in a mouse model

Myeloid cells from bone marrow were analyzed using anti-mouse CD370 (10B4) in flow cytometry. Viable cells were selected using propidium iodide exclusion. Expression changes were quantified under inflammatory conditions. CD370 MFI increased significantly in LPS-treated samples. Gating strategy focused on Ly6G+ and Ly6C+ subsets.

Flow cytometry-based detection of CD370 on dendritic cells with anti-Dectin-1 clone 10B4

Dendritic cells were stained with anti-Dectin-1 (clone 10B4) for flow cytometric quantification. Phagocytosis was assessed by uptake of fluorescent beads. BD LSR II was used for data acquisition. Mature DCs showed higher CD370 expression. Blocking with 10B4 reduced phagocytic activity.
