Background of Recombinant Anti-mouse CD370 Monoclonal Antibody (Clone: 10B4)

The rat anti-mouse CD370 (Clec9A or DNGR-1) monoclonal antibody 10B4 (rat IgG2a kappa) reacts with the mouse CD370 protein. CLEC9A (C-type lectin domain family 9 member A), a transmembrane glycoprotein with a single extracellular C-type lectin domain (CTLD), plays a role in the immune response. As an activation receptor, CLEC9A is expressed on myeloid lineage cells, helps process material from dead cells, recognizes filamentous actin when cell membranes are damaged, and helps present antigens to cytotoxic T cells. CLEC9A has an intracellular cytoplasmic tail with potential signaling motifs. CLEC9A is expressed on a subset of BDCA3+ dendritic cells and CD16- monocytes in humans, and is expressed on CD8+ conventional DCs and plasmacytoid DCs in mice.

The in vivo grade recombinant anti-mouse CD370 rat IgG2a monoclonal antibody was produced in mammalian cells. 
Clone: 10B4.
Isotype: Rat IgG2a kappa.
Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: 10B4) specifically binds to mouse CD370.
Applications: ELISA, flow cytometry, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse CD370 protein. 
Formulation: 0.2 μM filtered solution of 1x PBS.
Purity: >95% by SDS-PAGE under reducing conditions.
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.

The 10B4 system of human T cells seems to constitute a member of T cell receptor complex. ...The 10B4 antibody (bioactivity of anti-mouse CD370 10B4) together with goat anti-mouse immunoglobulin antibody induced significant increase of DNA and RNA syntheses in T cells with peak responses on day 9 and on day 7, respectively. ...Blocking studies with pharmacological agents showed that in the 10B4-induced system, a protein kinase C (PK-C) inhibitor, palmitoylcarnitine, blocked DNA synthesis, RNA synthesis, IL-2 production and IL-2R expression whereas a Ca ion channel blocker, verapamil, inhibited DNA synthesis, RNA synthesis, IL-2 production but not IL-2R expression. ...We have analyzed the mechanisms by which a monoclonal antibody against the 10B4 molecule (anti-mouse CD370 (10B4) in vivo antibody) activates human peripheral T cells. ...It is thus concluded that PK-C activation is required for IL-2 production and IL-2R expression and that channel-mediated Ca ion influx is important for IL-2 production but may not be needed for IL-2R expression.
