The rat anti-mouse CD370 (Clec9A or DNGR-1) monoclonal antibody 10B4 (rat IgG2a kappa) reacts with the mouse CD370 protein. CLEC9A (C-type lectin domain family 9 member A), a transmembrane glycoprotein with a single extracellular C-type lectin domain (CTLD), plays a role in the immune response. As an activation receptor, CLEC9A is expressed on myeloid lineage cells, helps process material from dead cells, recognizes filamentous actin when cell membranes are damaged, and helps present antigens to cytotoxic T cells. CLEC9A has an intracellular cytoplasmic tail with potential signaling motifs. CLEC9A is expressed on a subset of BDCA3+ dendritic cells and CD16- monocytes in humans, and is expressed on CD8+ conventional DCs and plasmacytoid DCs in mice.
Our recombinant 10B4 antibodies have a part (variable regions) or complete amino acid sequences of the rat anti-mouse CD370 monoclonal antibody (hybridoma clone name or number: 10B4).
The in vivo grade recombinant anti-mouse CD370 rat IgG2a monoclonal antibody was produced in mammalian cells.
Clone: 10B4.
Isotype: Rat IgG2a kappa.
Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: 10B4) specifically binds to mouse CD370.
Applications: ELISA, flow cytometry, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse CD370 protein.
Formulation: 0.2 μM filtered solution of 1x PBS.
Purity: >95% by SDS-PAGE under reducing conditions.
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.
Shipping: The in vivo grade recombinant anti-mouse CD370 antibodies (clone of 10B4) are shipped with ice pack. Upon receipt, store it immediately at the temperature recommended below.
Stability & Storage: Use a manual defrost freezer and avoid repeated freeze-thaw cycles.

Mice were normally injected intravenously with 10 μg of rat anti-Clec9A mAb (10B4) or isotype control mAb1 (IgG2a; eBioscience) or isotype control mAb2 (IgG2a, anti–β-gal; GL117). Serum samples were obtained at 2 to 8 weeks and the level of anti–rat Ig reactivity determined by enzyme-linked immunosorbent assay (ELISA)...The injection of the anti-mClec9A mAb 10B4 alone, without any additional DC activation agents, produced a strong and prolonged anti–rat Ig response (Figure 5).
