Description

Background

The in vivo grade recombinant anti-mouse CD25 monoclonal antibodies are based on the rat monoclonal antibody anti-CD25 (mouse) with the clone number of PC61, PC61.5 or PC61.5.3. The rat IgG1 lamda monoclonal antibody has been validated and reported for use in immunoprecipitation (IP), immunohistochemistry (IHC), Flow Cytometry (FC), and various functional assays, such as in vivo depletion of CD25+CD4+ Treg cells, in vitro blocking of IL-2 binding to low- and high-affinity receptors, and growth inhibition of IL-2-dependent T-cell lines. Expression of CD25, together with CD4 and FOXP3, is considered a phenotypic signature for Treg cells. In vivo depletion of mouse CD25(+)CD4(+)FoxP3(+) Treg cells using the rat PC61 monoclonal antibody is widely used to characterize the Treg function. The PC61 monoclonal antibody specifically binds to an epitope of CD25 which is distinct from the IL-2 binding site and from those recognized by the 3C7 and 7D4 monoclonal antibodies. By doing so, the antibody blocks binding of IL-2 to CD25.

The PC61 anti-CD25 monoclonal antibody (rat IgG1) was used to deplete CD4+ FOXP3+ regulatory T cells (Tregs) in vivo, mediated by FcγRIII+ phagocytes. Mice treated with PC61 variants showed differential effects on Treg depletion in spleen and lymph nodes, confirmed by flow cytometry. The study clarified that while CD25 blockade maintains immune homeostasis, active depletion of CD25+ Tregs leads to aberrant T-cell activation, highlighting distinct impacts of PC61 on Treg biology.

Role of CD25+ dendritic cells in the generation of Th17 autoreactive T cells in autoimmune experimental uveitis

The PC61 anti-CD25 antibody was administered in vivo to C57BL/6 mice immunized with the uveitogenic peptide interphotoreceptor retinoid-binding protein, reducing the Th17 response while enhancing the autoreactive Th1 response. Splenic T cells from PC61-treated or non-treated mice were enriched 13 days post-immunization and stimulated in vitro with IRBP1–20 peptide for 48 hours. The study analyzed the effects of PC61 on CD25+CD3+, Foxp3+, and γδ T cells, confirming its impact on T-cell phenotypes.

In vivo depletion of CD4+FOXP3+ Treg cells by the PC61 anti-CD25 monoclonal antibody is mediated by FcgammaRIII+ phagocytes

The PC61 monoclonal antibody (anti-mouse CD25, rat IgG1) was used to deplete CD4+CD25+FoxP3+ Treg cells in vivo, with a single injection eliminating approximately 70% of these cells in peripheral blood, while remaining Tregs showed low or no CD25 expression. The study revealed that Treg depletion is mediated by FcγRIII+ phagocytes, with clodronate liposome treatment inhibiting depletion, whereas NK cell depletion had no effect, elucidating the mechanism of PC61 action.

The PC61 monoclonal antibody was used for in vivo depletion of CD4+CD25+ regulatory T cells in athymic nude mice reconstituted with CD4+ T cells. Intraperitoneal injection of PC61 led to depletion of Tregs, which was necessary but not sufficient for inducing organ-specific autoimmune disease, as confirmed by histological analysis of target organs. Reconstitution with CD25-depleted T cells resulted in multi-organ autoimmunity.
