The QBEnd/10 monoclonal antibody detected CD34 antigen on hematopoietic cells via flow cytometry, adhering to ISHAGE guidelines. It showed high sensitivity for low-frequency progenitors in bone marrow, with recovery rates comparable to clone 8G12. Quality control used isotype controls and 7-AAD viability staining.

QBEnd/10 antibody, conjugated to PE, was used in a three-color flow cytometry protocol for CD34+ cell counting in umbilical cord blood analyzed within 24 hours. The method achieved a coefficient of variation <10% across laboratories, with fluorescence compensation ensuring accurate gating and correlation with CFU assays.

QBEnd/10-FITC, one of five anti-CD34 antibodies, was tested in flow cytometry on PBSC apheresis products using a FACSCalibur, showing superior specificity for dim CD34+ populations. Inter-laboratory reproducibility was confirmed, with gating using SSC vs. CD45 to exclude debris and lymphocytes.

The QBEnd/10 antibody was used in a standardized flow cytometry panel for AML immunophenotyping in bone marrow aspirates from 200 patients. Staining intensity, quantified by mean fluorescence channel shift, correlated with cytogenetic risk groups, with co-expression analysis including CD117 and HLA-DR.

The QBEnd/10 monoclonal antibody was validated for CD34+ cell enumeration in PBPC products, achieving an intra-assay CV of 5.2% at 1% positivity and linearity across 0.1–10% CD34+ frequencies. Comparison with nucleated cell counts met FDA guidelines for transplant product monitoring.
