Introduction
DARPP-32 is localized to neurons containing dopamine receptors and is a potent inhibitor of protein phosphatase 1, which plays a central role in dopaminergic and glutamatergic signaling, and in integrating the activity of these two pathways. Dopamine, acting through the D1 receptor and the adenylate cyclase-stimulatory G protein Gs/Golf, activates adenylate cyclase, thereby bringing about protein kinase A (PKA)-mediated phosphorylation of DARPP-32. By contrast, acting through the D2 receptor and the adenylate cyclase-inhibitory Gi proteins, dopamine reduces DARPP-32 phosphorylation. Calcineurin acts as a principle mediator of dephosphorylation-dependent inactivation of DARPP-32 (see ref. 5 for a review). Consistent with these observations, knockout mice with disruption of the DARPP-32 gene have been previously shown to have alterations in their molecular, electrophysiological, and behavioral responses to dopamine, drugs of abuse, and antipsychotic medications.

Specificity/Sensitivity: DARPP-32 (phospho-Thr75) antibody detects endogenous levels of DARPP-32 only when phosphorylated at threonine 75.

Immunogen: The antiserum was produced against synthesized phosphopeptide derived from human DARPP-32 around the phosphorylation site of threonine 75 (A-Y-TP-P-P).
Purification: The antibody was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific phosphopeptide. The antibody against non-phosphopeptide was removed by chromatography using non-phosphopeptide corresponding to the phosphorylation site.
Form of Antibody: Rabbit IgG in phosphate buffered saline (without Mg2+ and Ca2+ ), pH 7.4, 150 mM NaCl, 0.02% sodium azide and 50% glycerol.
Storage/Stability: Stable for 1 year at -20°C and 3 months at 4°C. For maximum recovery of the product, centrifuge the original vial after thawing and before removing the cap. Aliquot to avoid repeated freezing and thawing.
