Introduction
The Disabled-1 (Dab1) gene encodes a key regulator of Reelin signaling. Reelin is a large glycoprotein secreted by neurons of the developing brain, particularly Cajal-Retzius cells. DAB1 functions downstream of Reln in a signaling pathway that controls cell positioning in the developing brain and during adult neurogenesis. It docks to the intracellular part of the Reelin very low density lipoprotein receptor (VLDLR) and apoE receptor type 2 (ApoER2) and becomes tyrosine-phosphorylated following binding of Reelin to cortical neurons. In mice, mutations of Dab1 and Reelin generate identical phenotypes. In humans, Reelin mutations are associated with brain malformations and mental retardation. In mice, Dab1 mutation results in the scrambler mouse phenotype. With a genomic length of 1.1 Mbp for a coding region of 5.5 kb, DAB1 provides a rare example of genomic complexity, which will impede the identification of human mutations.
PA000436-B7055: Dab1 (Ab-232) Polyclonal Antibody
Rabbit Polyclonal Antibody.
Specificity/Sensitivity: Dab1 (Ab-232) antibody detects endogenous levels of total Dab1 protein.
Applications: WB: 1:500~1:1000 IF: 1:500~1:1000 ELISA: 1:40000.
Reactivity: Human, Mouse, Rat.
Immunogen: The antiserum was produced against synthesized non-phosphopeptide derived from human Dab1 around the phosphorylation site of tyrosine 232 (G-V-YP-D-V).
Purification: The antibody was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific immunogen.
Form of Antibody: Rabbit IgG in phosphate buffered saline (without Mg2+ and Ca2+), pH 7.4, 150 mM NaCl, 0.02% sodium azide and 50% glycerol.
Storage/Stability: Stable for 1 year at -20°C and 3 months at 4°C. For maximum recovery of the product, centrifuge the original vial after thawing and before removing the cap. Aliquot to avoid repeated freezing and thawing.
