Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: R2-9A5) specifically binds to mouse IL-12 p75.

The R2-9A5 monoclonal antibody, targeting the p75 subunit of IL-12, was administered intraperitoneally at 2 mg/kg every other day starting from the day of immunization, completely preventing experimental autoimmune encephalomyelitis (EAE) in SJL/J mice. Clinical signs of EAE were scored daily on a 0-5 scale, with treated mice showing reduced Th1 cytokine production in the CNS and decreased inflammation in spinal cord sections confirmed by histological examination.

The anti-mouse IL-12 p75 antibody R2-9A5 was injected intraperitoneally at 500 μg per mouse twice weekly, significantly reducing arthritis severity in DBA/1 mice induced by type II collagen in CFA. Joint swelling was measured using calipers every three days, and cytokine analysis revealed decreased IFN-γ and increased IL-4 levels, indicating a shift away from Th1 responses.

In vivo blockade of IL-12 p75 with R2-9A5 prevents allergic airway inflammation

The R2-9A5 monoclonal antibody, targeting mouse IL-12 p75, was administered subcutaneously at 100 μg per dose during sensitization, neutralizing IL-12 in vivo during OVA challenge in a mouse model of allergic airway inflammation. Airway hyperresponsiveness was assessed by methacholine challenge, and bronchoalveolar lavage (BAL) fluid analysis showed reduced eosinophil counts and goblet cell hyperplasia in airways of R2-9A5-treated mice.

R2-9A5-mediated neutralization of IL-12 p75 attenuates graft-versus-host disease

The anti-IL-12 p75 antibody R2-9A5 was administered intravenously at 1 mg/kg post-transplantation, reducing graft-versus-host disease (GVHD) mortality in a mouse bone marrow transplant model. GVHD scores were based on weight loss, activity, and fur texture, with histopathology showing decreased intestinal and liver damage. T cell activation was suppressed, as measured by flow cytometry, in R2-9A5-treated mice.

Role of IL-12 p75 in acute lung injury: blockade with R2-9A5 antibody

The anti-mouse IL-12 p75 antibody R2-9A5 was used to block IL-12 in a mouse model of acute lung injury induced by intratracheal LPS administration. Lung permeability was assessed by Evans blue dye extravasation, and neutrophil sequestration was quantified in bronchoalveolar lavage. R2-9A5 treatment reduced vascular leakage by 60% compared to controls, demonstrating its anti-inflammatory effects.
