Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: 20C2) specifically binds to human IL-12.
Applications:ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human IL-12 protein.

Targeting IL-12 family cytokines in inflammatory autoimmune diseases

The 20C2 antibody, administered intraperitoneally at 200 μg weekly, neutralized IL-12 p70 in vivo, significantly reducing Th1-mediated inflammation in the experimental autoimmune encephalomyelitis (EAE) model induced by MOG35-55 peptide in C57BL/6 mice. Clinical scores monitored daily showed decreased disease progression, and IFN-γ production in spleen cells was markedly reduced.

The 20C2 antibody was used to neutralize IL-12 p70 in C57BL/6 mice infected with 2 × 10^5 PFU of LCMV, resulting in a significantly reduced CD8+ T cell response. Serum cytokine levels, measured by ELISA 48 hours post-infection, confirmed the impact of IL-12 neutralization, while plasmacytoid dendritic cells were depleted using anti-mPDCA-1 antibody.

Endogenous IL-12 is not required for susceptibility to Trypanosoma cruzi infection

The 20C2 antibody neutralized endogenous IL-12 during Trypanosoma cruzi infection in mice, with parasitemia monitored by counting trypomastigotes in peripheral blood. Survival rates were recorded for 30 days, showing no significant difference in tissue parasite burden compared to IL-12 p40 knockout mice used as controls.

IL-12 is required for the development of experimental autoimmune encephalomyelitis

Neutralization of IL-12 with the 20C2 antibody completely prevented experimental autoimmune encephalomyelitis (EAE) development in mice immunized with 100 μg of MOG peptide in complete Freund’s adjuvant. Clinical symptoms (scored 0–5) and histological analysis of spinal cord sections showed reduced inflammatory cell infiltration in the CNS.

Role of endogenous IL-12 in the Th1 response to Leishmania major

Administration of the 20C2 antibody to neutralize IL-12 in C57BL/6 mice infected with 2 × 10^5 Leishmania major promastigotes increased lesion size. Lymph node cells showed impaired Th1 development, and parasite burden was quantified by limiting dilution analysis, confirming the critical role of IL-12 in the Th1 response.
