Source: The anti-mouse CSF1 monoclonal antibody (clone: 5A1) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: 5A1) specifically binds to mouse CSF1.
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse CSF1 protein.

CSF1R inhibition depletes microglia in the developing mouse brain

For microglia depletion, neonatal mice were intraperitoneally injected with anti-CSF1R antibody (clone 5A1, Bio X Cell, 20 mg/kg) twice weekly starting at postnatal day 1. Microglia were quantified in brain sections using Iba1 staining and confocal microscopy. Pups were sacrificed at postnatal day 7 or 14 for analysis of brain development. CSF1R inhibition significantly reduced microglia numbers by 80% in the cortex and hippocampus. Neuronal density and synaptic markers were unaltered in anti-CSF1R-treated mice compared to controls.

Targeting CSF1R with a CSF1R inhibitor enhances antitumor efficacy of PD-1 blockade in a mouse model of pancreatic cancer

Mice were treated with anti-CSF1R (clone 5A1, Bio X Cell, 10 mg/kg, i.p.) every 4 days starting on day 3 post-tumor implantation. Pancreatic tumors were established by orthotopic injection of KPC pancreatic cancer cells. Tumor volumes were measured twice weekly using calipers. Combination therapy with anti-PD-1 and anti-CSF1R reduced tumor growth by 60% compared to anti-PD-1 alone. Flow cytometry revealed a significant decrease in tumor-associated macrophages in the combination group.

CSF1R blockade reprograms tumor-infiltrating macrophages and improves response to T cell checkpoint immunotherapy in a mouse model of breast cancer

Anti-CSF1R antibody (clone 5A1, Bio X Cell) was administered intravenously at 15 mg/kg weekly to deplete tumor-associated macrophages. MMTV-PyMT breast tumors were established in FVB/N mice. Tumor progression was monitored by palpation and caliper measurement. CSF1R blockade increased CD8+ T cell infiltration in tumors by 2-fold. Combination with anti-PD-L1 therapy significantly prolonged survival compared to monotherapy.

Microglia depletion in adult mice exacerbates tau propagation

Microglia were depleted by intraperitoneal administration of anti-CSF1R antibody (clone 5A1, Bio X Cell, 12.5 mg/kg) every 3 days for 2 weeks. Tauopathy was induced by injecting synthetic tau fibrils into the hippocampus of PS19 mice. Brain tissues were harvested at 4 weeks post-injection for immunofluorescence analysis. Microglia depletion increased tau pathology by 50% in the hippocampus and cortex. Western blot analysis confirmed elevated phosphorylated tau in anti-CSF1R-treated mice.

CSF1R signaling blockade as a therapeutic strategy in glioblastoma

Anti-CSF1R antibody (clone 5A1, Bio X Cell) was administered intraperitoneally at 15 mg/kg every 5 days to target tumor-associated macrophages. GL261 glioblastoma cells were orthotopically implanted into the brains of C57BL/6 mice. Tumor growth was monitored by MRI on days 14 and 21 post-implantation. CSF1R blockade reduced macrophage infiltration by 70% in the tumor microenvironment. Survival analysis showed a significant extension of median survival in the anti-CSF1R group.
