Mice were injected intraperitoneally with 200 μg of anti-mouse CD209b antibody (clone 22D1, Bio X Cell) 1 hour prior to CLP surgery. Sepsis was induced by cecal ligation and puncture (CLP). Survival was monitored for 96 hours post-CLP. 22D1 treatment increased survival rate to 70%. Bacterial loads in blood were reduced by 2 logs.

In vivo neutralization of CD209b using clone 22D1 attenuates allergic airway inflammation

Anti-mouse CD209b antibody (clone 22D1, BioLegend) was administered intravenously at 150 μg per dose weekly for 4 weeks during OVA challenge. Allergic airway inflammation was induced by OVA sensitization and aerosol challenge. Airway hyperresponsiveness was assessed by methacholine provocation. CD209b blockade reduced eosinophil infiltration by 50%. Th2 cytokines in BAL fluid were quantified by ELISA.

Role of SIGN-R1 blockade with 22D1 in experimental autoimmune encephalitis

Mice received 100 μg of anti-mouse CD209b antibody (clone 22D1, Bio X Cell) intraperitoneally daily for 10 days starting at EAE induction. EAE was induced by MOG35-55 peptide immunization in CFA. Clinical scores were recorded daily. 22D1 treatment lowered mean clinical score by 3 points. CNS inflammation was evaluated by histology.

Anti-CD209b therapy with 22D1 modulates dendritic cell function in a viral infection model

Anti-mouse CD209b antibody (clone 22D1, Bio X Cell) was injected intravenously at 250 μg 24 hours prior to viral challenge. Viral infection was established by intranasal LCMV administration. Viral titers in lung were measured by plaque assay on day 5. CD209b blockade enhanced DC maturation. T cell priming was assessed in draining lymph nodes.

Blockade of CD209b with clone 22D1 in a tumor metastasis model

Mice were treated with 300 μg of anti-mouse CD209b antibody (clone 22D1, BioLegend) intraperitoneally every 3 days for 2 weeks post-tumor injection. B16 melanoma cells were injected intravenously to induce lung metastasis. Metastatic foci were counted in lung sections on day 14. 22D1 reduced metastatic burden by 40%. Tumor cell adhesion to endothelium was inhibited in vivo.
